AB  - In this study, levan from Bacillus licheniformis NS032 was modified in an aqueous medium by octenyl succinic
anhydride (OSA), and the properties of the obtained derivatives were studied. The maximum efficiency in the
synthesis reaction was achieved at 40 ◦C and a polysaccharide slurry concentration of 30 %. Increasing the
reagent concentration (2–10 %) led to an increase in the degree of substitution (0.016–0.048). Structures of
derivatives were confirmed by FTIR and NMR. Scanning electronic microscopy, thermogravimetry, and dynamic
light scattering analyses showed that the derivatives with degrees of substitution of 0.025 and 0.036 retained
levan’s porous structure and thermostability and showed better colloidal stability than the native polysaccharide.
The intrinsic viscosity of derivatives increased upon modification, while the surface tension of the 1 % solution
was lowered to 61 mN/m. Oil-in-water emulsions prepared with sunflower oil (10 % and 20 %) by mechanical
homogenization and 2 and 10 % derivatives in the continuous phase showed mean oil droplet sizes of 106–195
μm, while the distribution curves exhibited bimodal character. The studied derivatives have a good capacity to
stabilize emulsions, as they have a creaming index ranging from 73 % to 94 %. The OSA-modified levans could
have potential applications in new formulations of emulsion-based systems.

Mushroom β-glucan and polyphenol formulations as natural immunity boosters and balancers: nature of the application

AB  - Mushrooms are experiencing a kind of renaissance as a part of the contemporary human diet. These valuable organisms are more than food, they fi t in perfectly as a novel market group known as nutra-mycoceuticals. Immune-balancing mushroom dietary fibers and secondary metabolites such as polyphenols are the main focus of the healthcare industry. Wellness and cosmetic companies are increasingly using mushroom extracts rich in these ingredients. This review considers the basic molecular immunomodulatory mechanisms of action of the most commonly used mushroom dietary fibers, β-glucans. The literature data on their bioavailability, metabolic transformations, preclinical and human clinical research, and safety are discussed. Immunomodulatory mechanisms of polyphenol ingredients are also considered. These molecules present great potential in the design of the new immunity balancer formulations according to their widespread structural diversity. Finally, we draw attention to the perspectives of modern trends in mushroom nutraceutical and cosmeceutical formulations to strengthen and balance immunity.

AB  - Excessive production of reactive oxygen species (ROS) often results in irreversible cell damage and leads to a number of disorders such as hypertension, heart disease, diabetes, inflammation, premature aging and cancer. Considering the importance of diet in prevention of diseases associated with oxidative stress, this study was conducted to evaluate antioxidant activity of the methanol extract of wild lignicolus mushroom Fomitopsis pinicola as natural source of functional food ingredients. Nowadays functional food products based on the lignicolous mushroom species are increasingly available on the market. Their powders and extracts are consumed as dietary supplements in the form of capsules or tablets and as additives in the formulation of healthier food products.

AB  - Aquaphotomics is a relatively novel scientific discipline which can be used for elucidation of structural and  related functional properties of aqueous systems. The aim of this study was to analyse water solutions of carbohydrates of different size from monosaccharides (glucose and fructose) via, disaccharides (sucrose), gluco and fructo-oligosaccharides and various selected polisaccharides (based on glucose and fructose, respectively) using Aquaphotomics. Knowing that main functions of carbohydrates within our body are various: providing energy and regulation of blood glucose; sparing the use of proteins for energy; breakdown of fatty acids and preventing ketosis; biological recognition processes; flavor and sweeteners and dietary fibers who can promote digestive health, knowledge about their interaction with water molecules can have strong influence on our understanding of some basic but also complex metabolic processes.

AB  - In recent years, fungi have been recognized as producers of acetylcholinesterase (AChE) inhibitors, agents important for the prevention of Alzheimer’s disease (AD). This study aimed to examine the AChE inhibitory, the antioxidative and antibacterial activity of two different Schizophyllum commune strains that originated from Serbia (SRB) and Italy (IT). Submerged cultivation of grown mycelia (M) and fermentation broth (F) of ethanol (EtOH) and polysaccharide (PSH) extracts lasted for 7, 14, 21 and 28 days. For AChE activity Ellman method was performed, while for antioxidative activity, sevendifferent assays were conducted: DPPH, ABTS, FRAP, SOA, OH, NO together with total phenolic content. Antimicrobial screen, LC–MS/MS technique and FTIR measurements were performed. Different isolates exhibited different AChE activity, with PSH being the strongest (SRB, M, 28 days IC90 79.73 ± 26.34 µg/mL), while in EtOH extracts, IT stood out (F, 14 days, IC50 0.8 ± 0.6 µg/mL). PSH extracts (7 days) exhibit significant antioxidative activity (AO), opposite to EtOH extracts where 14 and 21days periods stood out. Only tw extracts showed antibacterial activity. Following LC–MS/MS analysis p-hydroxybenzoic and gallic acids were the most abundant phenolics. PSH extracts demonstrated remarkable results, making this study debut and introducing S. commune as a valuable resource of AChE inhibitors.

abstract = "In recent years, fungi have been recognized as producers of acetylcholinesterase (AChE) inhibitors, agents important for the prevention of Alzheimer’s disease (AD). This study aimed to examine the AChE inhibitory, the antioxidative and antibacterial activity of two different Schizophyllum commune strains that originated from Serbia (SRB) and Italy (IT). Submerged cultivation of grown mycelia (M) and fermentation broth (F) of ethanol (EtOH) and polysaccharide (PSH) extracts lasted for 7, 14, 21 and 28 days. For AChE activity Ellman method was performed, while for antioxidative activity, sevendifferent assays were conducted: DPPH, ABTS, FRAP, SOA, OH, NO together with total phenolic content. Antimicrobial screen, LC–MS/MS technique and FTIR measurements were performed. Different isolates exhibited different AChE activity, with PSH being the strongest (SRB, M, 28 days IC90 79.73 ± 26.34 µg/mL), while in EtOH extracts, IT stood out (F, 14 days, IC50 0.8 ± 0.6 µg/mL). PSH extracts (7 days) exhibit significant antioxidative activity (AO), opposite to EtOH extracts where 14 and 21days periods stood out. Only tw extracts showed antibacterial activity. Following LC–MS/MS analysis p-hydroxybenzoic and gallic acids were the most abundant phenolics. PSH extracts demonstrated remarkable results, making this study debut and introducing S. commune as a valuable resource of AChE inhibitors.",

AB  - Essential oils extracted from plants are attractive additives in the food industry due to their antimicrobial and antioxidant properties. Therefore, in recent years, essential oils have been extensively investigated as additives for syntheses of edible and biodegradable emulsified films and coatings. Edible films and coatings are used in food preservation in order to improve the gas and moisture barriers, mechanical properties, sensory perceptions, microbial protection and to 
prolong the shelf life of various food products. Finally, edible films should replace conventional materials originating from petrochemical industry. The materials for the production of edible films include mainly proteins such as gelatin, lipids, and polysaccharides individually or in combination. Sometimes additives are incorporated to improve the properties of films. Microbial levan is a biocompatible, biodegradable and a renewable exopolysaccharide with antioxidant and prebiotic activities.

AB  - Total quality index approach was used to reflect the quality of Pleurotus ostreatus mushroom, cultivated on grape pomace (P) as food waste substrate and wheat straw (S), in four different combined ratios: 100P; 80P20S; 50P50S; and 20P80S, with 100S, serving as control substrate. This approach involved two directions of mushroom analysis. The whole mushroom fruiting bodies analysis introduced a single total quality index (TQIM) based on 11 quality parameters. The mushroom water extracts analysis was presented by another single total quality index (TQIE), with seven quality parameters. This mathematical model revealed that the mushroom extracts have a better overall quality score with higher pomace content in substrate mixture, compared to the whole mushroom quality changes throughout the observed shelf life period. Future research may pay attention to the phytotoxicity of grape pomace as a food waste, as well as the influence of grape pomace on glucan content, as a health‐beneficial compounds.

AB  - Ongoing demand in sustainable and biocompatible drug dosage forms is reflected in the search for novel pharmaceutical excipients with equal properties. A group of microbial exopolysaccharides offers a variety of biopolymers with many alleged uses and effects. This study aims to assess applicative properties of levan obtained from Bacillus licheniformis NS032, focusing on its potential co-stabilizing and drug release-controlling functions in pertaining emulsion systems. Despite its high molecular weight and partial existence in globular nanometric structures (180-190 nm), levan was successfully incorporated into both tested colloidal systems: those stabilized with synthetic/anionic or natural-origin/non-ionic emulsifiers. In the tested levan concentrations range (0.2-3.0% w/w) the monitored flow and thermal parameters failed to show linear concentration dependence, which prompted us to revisit certain colloidal fundamentals of this biopolymer. Being a part of the external phase of the investigated emulsion systems, levan contributed to formation of a matrix-like environment, offering additional stabilization of the microstructure and rheology modifying properties (hysteresis loop elevation as high as 4167±98 to 20792±3166 Pa•s−1), especially in case of the samples where lamellar liquid crystalline formation occurred. Apart from its good water solubility and considerable conformational flexibility, the investigated homofructan easily saturated the external phase of the samples stabilized with a conventional anionic emulsifier, leading to similar properties of 0.2% and 3.0% levan-containing samples. After closer consideration of thermal and release behavior, this was considered as a favorable property for a novel excipient, offering tailored formulation characteristics even with lower levan concentrations, consequently not compromising the potential cost of the final drug dosage form.

AB  - An efficient encapsulation of the fullerene into two hydrophobized and one native polysaccharide provided water soluble supramolecular hybrids. After covalent modification of polysaccharides by cholesterol, noncovalent hybrids were prepared by a three-step procedure, including mixing of individual aqueous solutions of hydrophobized, as well as native sugar with solution of the fullerene in pyridine, dialysis and lyophilization. Although the degree of the fullerene incorporation into hydrophobized substrates, cholesterol-levan and cholesterol-pullulan, was lower in comparison to the native polysaccharide levan, hydrophobization provided nanoparticles with improved properties. The particle size distribution, studied by dynamic light scattering and scanning electron microscopy revealed formation of moderately polydisperse aggregates, with the diameter contraction in comparison to the corresponding free polysaccharide, especially in the case of hydrophobized substrates. The morphological examination, done by scanning electron microscopy indicated the self-organization of the fullerene-native polysaccharide to round individual structures, while fullerene-hydrophobized polysaccharide hybrids tend to form networks. The antioxidant activity of the synthesized polysaccharide-C60 noncovalent hybrids versus starting polysaccharides was investigated by the DPPH radical scavenging and the β-carotene-linoleic acid bleaching methods. In all three complexes, the radical scavenging ability of the fullerene remained preserved, and a positive effect of levan hydrophobization was observed.

AB  - An efficient encapsulation of the fullerene into two hydrophobized and one native polysaccharide provided water soluble supramolecular hybrids. After covalent modification of polysaccharides by cholesterol, noncovalent hybrids were prepared by a three-step procedure, including mixing of individual aqueous solutions of hydrophobized, as well as native sugar with solution of the fullerene in pyridine, dialysis and lyophilization. Although the degree of the fullerene incorporation into hydrophobized substrates, cholesterol-levan and cholesterol-pullulan, was lower in comparison to the native polysaccharide levan, hydrophobization provided nanoparticles with improved properties. The particle size distribution, studied by dynamic light scattering and scanning electron microscopy revealed formation of moderately polydisperse aggregates, with the diameter contraction in comparison to the corresponding free polysaccharide, especially in the case of hydrophobized substrates. The morphological examination, done by scanning electron microscopy indicated the self-organization of the fullerene-native polysaccharide to round individual structures, while fullerene-hydrophobized polysaccharide hybrids tend to form networks. The antioxidant activity of the synthesized polysaccharide-C60 noncovalent hybrids versus starting polysaccharides was investigated by the DPPH radical scavenging and the β-carotene-linoleic acid bleaching methods. In all three complexes, the radical scavenging ability of the fullerene remained preserved, and a positive effect of levan hydrophobization was observed.

AB  - The health promoting effects of hot water extracts obtained from
fruiting bodies of the commercially cultivated mushroom Agaricus bisporus
(AbHW) and the wild-growing mushroom Ganoderma resinaceum (GrHW)
originating from northern Serbia are presented in this research. These abilities
were compared in vitro by the prevention of lipid peroxidation (LPx) in a linoleic
acid model system, inhibition of the angiotension converting I enzyme
(ACE) that could help in the maintenance of a normal blood pressure level and
strengthening the ability of the central cholinergic neuron by inhibiting the activity
of acetylcholinesterase (AChE). Cytotoxic activities were observed
towards selected human malignant (HeLa and K562) cell lines and normal-
-human peripheral blood mononuclear cells (PBMC). GrHW contains higher
phenolics (5.9 g (100 g)-1), inhibition of LPx (EC50 = 1.07 mg mL-1), ACE
(IC50 = 0.54 mg mL-1) and AChE (IC50 = 0.37 mg mL-1), and exhibited a significant
selectivity in the antitumour action against HeLa (IC50 = 0.14 mg mL-1)
and K562 (IC50 = 0.11 mg mL-1) cells. AbHW contained higher total protein
(6.4 g (100 g)-1), carbohydrate (75.4 g (100 g)-1) and β-glucan (55.1 g (100 g)-1)
contents and induced significant proliferation of healthy PBMC from 152–116
% in the concentration range of 0.047–0.187 mg mL-1. The difference in the
biological activity of the extracts provides guidance on their use as functional
food.
AB  - У овоме раду поређен је здраствени ефекaт врелих водених екстраката добијених из плодоносних тела комерцијално узгајане јестиве гљиве Agaricus bisporus (AbHW) и самоникле врсте гљиве Ganoderma resinaceum (GrHW) из региона северне Србије. Здраствени ефекат је поређен in vitro превенцијом липидне пероксидације (LPx) у модел систему линолеинске киселине, инхибицијом ангиотензин конвертујућег ензима (ACE) који има улогу у одржавању нормалног нивоа крвног притиска и јачањем способности централних холинергичких неурона инхибицијом активности ацетилхолинестеразе (AChE).
Цитотоксична активност је праћена на хуманим ћелијама тумора грлића материц (HeLa) и ћелијама хроничне мијелоидне леукемије (K562), као и на здравим мононуклеарним ћелијама периферног крвотока (PBMC). GrHW је показао већи садржај фенолних компоненти (5,9 g (100 g)-1), већу способност инхибиције LPx (EC50 = 1,07 mg mL-1),

AB  - Food packaging concerns a preservation and protection of all types of food and therefore is important part of food technology. Today, synthetic packaging films are predominantly used, however they are very slow or only partially degradable. Due to incomplete degradation of synthetic polymers, micro and nanoplastic could be produced, which is in last year’s considered as a new threat to human health. Microbial polysaccharide levan is natural biopolymer produced by broad range of microorganisms and has promising properties such as biocompatibility, renewability, high molecular height, low viscous nature, antioxidant and prebiotic activities. Gelatine, due to its affordability, biodegradability, low cost and excellent functional and film properties is currently the most preferred protein derivative.

AB  - Microbial polysaccharides, due to their unique physiochemical properties, have found application in the food industry, cosmetics, pharmacy and medicine. In the environment, microbes can use polysaccharides to alleviate the adverse effects of heavy metals in their close proximity. This adaptive property shows interesting potential for bioremediation. Herein, the effects of the exopolysaccharides (EPS) levan, produced by the bacterium Bacillus licheniformis NS032 and pullulan, produced by the fungus Aureobasidium pullulans CH-1 in the presence of copper (Cu2+) have been investigated for the first time on antioxidant enzyme activity, respiration and Cu2+ bioaccumulation of Daphnia magna as well as the bioluminescence of Vibrio fischeri. Both EPS decreased toxicity of Cu2+ in the acute test with D. magna. The activity of catalase (CAT) was significantly diminished after acute exposure to Cu2+ in comparison to treatments with Cu2+ and EPS, while in the prolonged acute exposure the CAT activity did not show statistically significant (P ≤ 0.05) differences between treatments with and without the EPS. According to ICP-MS results, during prolonged acute exposure of neonates, the bioaccumulation of Cu2+ in treatments without the EPS was 52.03 μg/g of biomass (wet), while in treatments with EPS, the bioaccumulation was lower by one order of magnitude. The respiration of neonates during acute exposure to Cu2+ with or without the EPS was monitored using the MicroOxymax respirometer, and the results show the EPS can positively effect the respiration. In the case of bacterial bioluminescence, the toxicity of Cu2+ decreased in treatments with EPS (30 min EC10) from 3.54 mg/L to 140.61 mg/L (levan) and 45.00 mg/L (pullulan). This study demonstrates protective effect of EPS against Cu2+ toxicity on D. magna and V. fischeri, and opens the door for further investigation of potential application of levan and pullulan in bioremediation of heavy metals and mitigation of their adverse effects in the aquatic environment.

High polysaccharide yields were produced by using diluted molasses (100-140 g/L of total sugars) with the addition of commercial sucrose up to 200 g/L of total sugars, as well as K2HPO4. A levan yield of 532 g/L was obtained on a medium optimized by response surface methodology, containing 62.6% of sugar originating from molasses, and 4.66 g/L of phosphate, with initial pH value of 7.2. In comparison to the media with 200 and 400 g/L sucrose, in the molasses optimized medium, the observed bacterial growth was faster, while the maximum production of polysaccharide was achieved over a shorter time interval (48 h). The polysaccharide produced in molasses medium had a weight average molecular weight of 5.82 x 10(6) Da, degree of branching 12.68%, viscosity of 0.24 dL/g, and based on methylation analysis and NMR data, it did not significantly differ from levan obtained in the medium with 200 g/L sucrose.

AB  - Three different polysaccharide extracts of a wild European source of the mushroom Ganoderma lucidum were screened for their free radical-blocking potential, which could strengthen the skin’s barrier function, and provide a skinlightening effect via potential inhibition of tyrosinase. The anti-collagenase and anti-elastase activities, which can help to restore skin elasticity and tensile strength, were also evaluated for the three extracts. Carbohydrates were the most abundant
components of the extracts, followed by smaller quantities of proteins, phenols and flavonoids. The glucan fraction represented between 48% and 61% of carbohydrate content in all three extracts. None of the analyzed extracts showed overt toxicity to spontaneously-transformed immortal human keratinocytes (HaCaT) at concentrations up to 2 mg/mL, and displayed superior scavenging ability on 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The hot water crude polysaccharide extract (HWCP) and partially purified fraction
(HWPP) were found to be effective inhibitors of lipid peroxidation (LPx), with an almost two-fold increased inhibition of LPx compared with ascorbic acid (EC50=1.65±0.08 mg/mL), a common additive in cosmeceutical formulations and used at mg levels. Among the investigated extracts, HWCP showed the strongest inhibition potential on tyrosinase and skin extracellular matrix (ECM) degradation enzymes. These diverse functionalities indicate that G. lucidum may represent a promising source of natural cosmeceutical ingredients.

AB  - In this work, pullulan based films with water extracts of medicinal herbs thyme and sage, were prepared. The resulting films were characterized by FT-IR spectroscopy and their antioxidant and antimicrobial activities were evaluated. AB  - The numerous naturally occurring biopolymeric materials are used as edible films and coatings (lipids, proteins, and polysaccharides). Microbial levan and pullulan have been already considered as biopolymers in food industry. The aim of this work was to obtain the levan and levan/pullulan blend films and compare them using the Atomic force microscopy (AFM) and Fourier transform infrared spectroscopy (FTIR). Two different films compositions were prepared by casting method: levan and levan/pullulan in the proportions 1:1. AFM measurements showed that levan film had a smoother surface and, consequently, a lower degree of roughness comparing to the levan/pullulan blend films. FTIR spectra showed peaks, which corresponded to all components of the obtained films.
