Two new jatrophane diterpenes from the roots of Euphorbia nicaeensis

AB  - In the previous study fifteen jatrophane diterpenes were isolated from the Euphorbia nicaeensis latex. Fourteen of them have been shown to be potent P-glycoprotein (P-gp) inhibitor in two MDR cancer cells (NCI-H460/R and DLD1-TxR). The aim of this study was to determine whether and which jatro­phane diterpenes can be isolated from the root of the plant, and then to examine their inhibition power on P-glycoprotein of selected cancer cell lines (NCI-H460, DLD1, U87, NCI-H460/R, DLD1-TxR and U87-TxR). Two previously undes­cribed jatrophane diterpenes were isolated from the root of E. nicaeensis col­lected in Deliblato Sand (Serbia). The structures of the isolated compounds were determined using 1D and 2D NMR, as well as HRESIMS data. The results obtained by MTT assay showed different antitumor potential of these two jatrophanes. Compound 1 inhibited cell growth of non-small cell lung car­cinoma cell lines NCI-H460 and NCI-H460/R, as well as glioblastoma cell lines U87 and U87-TxR, while jatrophane 2 was almost completely inactive in the suppression of cancer cell growth in a given range of concentrations. The obtained results also showed that the isolated compounds have an inhibitory effect on P-glycoprotein, as well as that their inhibitory potential is similar.
AB  - У претходном истраживању, петнаест дитерпена јатрофанског типа изоловано је из латекса Euphorbia nicaeensis. Њих четрнаест показала су се као снажни инхибитори P-гликопротеина (P-gp) у две MDR ћелијске линије рака (NCI-H460/R и DLD1-TxR). Циљ ове студије био је да се утврди да ли је и које јатрофанске дитерпене могуће изо- ловати из корена биљке, а затим испитивање њихове инхибиторне моћи на P-гликопро- теину одабраних ћелијских линија рака (NCI-H460, DLD1, U87, NCI-H460/R, DLD1-TxR и U87-TxR). Два претходно непозната јатрофана изолованa су из корена E. nicaeensis прикупљеног у Делиблатској пешчари. Структуре изолованих једињења одређене су применом 1D и 2D NMR метода, као и HRESIMS експеримента. Резултати добијени МТТ тестом показали су различит антиканцерогени потенцијал ова два јатрофана. Једињење 1 је инхибирало раст ћелија ћелијских линија неситноћелијског карцинома плућа NCI-H460 и NCI-H460/R, као и ћелијских линија глиобластома U87 и U87-TxR, док је јатрофан 2 био готово потпуно неефикасан у сузбијању растa ћелија карцинома у датом концентрационом опсегу. Добијени резултати су такође показали да 1 и 2 имају инхибиторно дејство на P-гликопротеин, као и да је њихов инхибиторни потенцијал сличан.

AB  - The presence of n-alkanes, free alcohols and free acids in leaf epicuticular wax extracts of 22 samples of 11 Euphorbia L. species belonging to the sections Paralias, Esula, Myrsiniteae and Helioscopia, 10 of which were never examined before, were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS), and n-alkane C27 was detected as the principal component of leaf epicuticular waxes in the majority of the examined species, while the most abundant free alcohol was C26. Three Euphorbia species belonging to section Helioscopia were characterized by a predominance of alcohol C28. Free acid (C16) was the major component in 21 investigated samples. The usefulness of n-alkanes and free alcohols and free acids as potential chemotaxonomic markers is briefly discussed.

Jatrophane Diterpenoids With Protective Effect on Human Lymphocytes DNA

AB  - Two sets of structurally different jatrophanes (1-11 and 13-16), jatrophane 12, and latex extract of 2 Euphorbia species (17 and 18) were tested for in vitro protective effect against chromosome aberrations in peripheral human lymphocytes using the cytokinesis-block micronucleus (CBMN) assay. Jatrophanes 1-6 in minimal doses of 1 µg/mL prominently decreased micronuclei (MN) frequency in the range 44.86% to 34.29% and manifested considerable protective effect. From the other set of jatrophanes, 13 in the same minimal dose notably decreased MN frequency by 31.05%, while extracts 17 and 18 at a concentration of 4 µg/mL remarkably decreased the frequency of MN by 37.94% and 36.12%, respectively. Jatrophanes 12, 14, and 16 showed moderate protection, while 7-11 and 15 were less active than positive control. The structure-activity relationship (SAR) studies of the tested jatrophanes (1-16) indicated the favorable position of benzoate at C-8 or C-9 (3, 4, and 13) and a preference of isobutanoyloxy group at C-3 (1-3) rather than propanoyloxy at the same position (4-6) for pronounced protective effect on human lymphocytes DNA. In a previous SAR study on 11 jatrophanes (1, 3-8, and 13-16), the same structural features in 3, 4, and 13 influenced powerful inhibition of P-gp, while growth inhibition of cancer cells was more than doubled in 1 (isobutanoyloxy group at C-3) compared to 6 (propanoyloxy at C-3).

The chemical structures of the compounds were determined by spectro-scopic analysis including 1D and 2D NMR and HRESIMS experiments. All but one of the previously undescribed jatrophanes, showed significant potential to inhibit P-glycoprotein (P-gp) activity in two MDR cancer cells (NCI-H460/12 and DLD1-TxR). The most powerful were nicaeenin F and nicaeenin G. Moreover nicaeenin G significantly stronger sensitized NCI-H460/R cells to DOX than Dex-VER.

Application of Spectroscopic Methods and Hyphenated Techniques to the Analysis of Complex Plant Extracts

AB  - Through the selected examples of different classes of secondary metabolites two general approaches in the analysis of complex mixtures of natural products are presented. The first one involves isolation of the constituents using optimized semi-prep LC in combination with classical column chromatography (CC) and the second one represents direct (on-line) analysis of complex plant extracts prior to isolation of the constituents. Thus the isolation of mg-quantities of jatrophanes the bioactive constituents of Euphorbia dendroides, with their structure determination on the basis of spectroscopic data interpretation (IR, NMR, MS), is described. The direct analysis of the plant extracts is demonstrated by the characterization of antioxidative polyphenols, i.e. oligomeric proanthocynidins from the MeOH extracts of grape Vitis vinifera seeds by the application of hyphenated methods (LC/UV/MS) in combination with 13C NMR spectroscopy, as well as tandem mass spectrometry (MS/MS) and MSn in combination with and without LC for identification of oligomeric proanthocynidins from other sources. The potential of another hyphenated instrumentation, LC/UV/SPE-NMR is shown by on-line identification of the various constituents of the crude extract of Hypericum perforatum. The possibilities and limitations of the NMR technique named diffusion ordered spectroscopy (DOSY) for direct analysis of the complex mixture, such as e.g. methanolic extract of Gentiana lutea, is discussed.

The direct analysis of the plant extracts is demonstrated by the characterization of antioxidative polyphenols, i.e. oligomeric proanthocynidins from the MeOH extracts of grape Vitis vinifera seeds by the application of hyphenated methods (LC/UV/MS) in combination with 13C NMR spectroscopy, as well as tandem mass spectrometry (MS/MS) and MSn in combination with and without LC for identification of oligomeric proanthocynidins from other sources. The potential of another hyphenated instrumentation, LC/UV/SPE-NMR is shown by on-line identification of the various constituents of the crude extract of Hypericum perforatum. The possibilities and limitations of the NMR technique named diffusion ordered spectroscopy (DOSY) for direct analysis of the complex mixture, such as e.g. methanolic extract of Gentiana lutea, is discussed.",

AB  - An ultra-performance liquid chromatographic triple quadrupole mass spectrometric (UPLC-QqQ-MS/MS) method with a multiple reactions monitoring mode (MRM) was developed and validated for the quantification of vanillin and syringaldehyde in plum brandy. The method showed good linearity (0.05 to 10 mg L-1) and low limits of detection and quantification (the LOD and LOQ values were 11.6 and 38.2 mu g L-1 for vanillin, and 12.7 and 42.0 mu g L-1 for syringaldehyde, respectively). The overall intra-day and inter-day variations were less than 4.21 % and the overall recovery was over 93.0 %. The correlation coefficients (R-2) of the calibration curves were higher than 0.9999. In order to evaluate whether the method was suitable for use as a routine analytical tool, vanillin and syringaldehyde were determined in 31 samples of Serbian plum brandy.

AB  - Dimers tomoroside A (1) and tomoroside B (2) of the co-occuring known chalcone monomer (3), along with the seven known flavonoid glucosides (4-10), were isolated from the aerial parts of Helichrysum zivojinii Cernjavski & Soska. The structures of the isolated compounds were elucidated by spectroscopic techniques. Compound 1 inhibited topo Ha and hif-1 alpha expression and stimulated doxorubicin anticancer effect, while 2 increased the expression of hif-1 alpha a, probably acting as antioxidant and redox status modulator. Notably, 2 synergized with Tipifarnib showing potential to improve the action of this new chemotherapeutic involved in the modulation of mitogene activated protein (MAP) kinase signaling pathway.

The structures of the isolated compounds were determined by UV, H-1-NMR and C-13-NMR spectroscopy and HR-ESI-MS spectrometry. H-1-NMR spectroscopy was used as a method for the quantitative analysis of cnicin.

AB  - Background: The aim of this research was to determine the intensity and mechanisms of the cytotoxic actions of five extracts isolated from the endemic plant species Helichrysum zivojinii Cernjavski & Soska (family Asteraceae) against specific cancer cell lines. In order to evaluate the sensitivity of normal immunocompetent cells implicated in the antitumor immune response, the cytotoxicity of extracts was also tested against healthy peripheral blood mononuclear cells (PBMC). Methods: The aerial parts of the plants were air-dried, powdered, and successively extracted with solvents of increasing polarity to obtain hexane, dichloromethane, ethyl-acetate, n-butanol and methanol extracts. The cytotoxic activities of the extracts against human cervix adenocarcinoma HeLa, human melanoma Fem-x, human myelogenous leukemia K562, human breast adenocarcinoma MDA-MB-361 cells and PBMC were evaluated by the MTT test. The mode of HeLa cell death was investigated by morphological analysis. Changes in the cell cycle of HeLa cells treated with the extracts were analyzed by flow cytometry. The apoptotic mechanisms induced by the tested extracts were determined using specific caspase inhibitors. Results: The investigated Helichrysum zivojinii extracts exerted selective dose-dependent cytotoxic actions against selected cancer cell lines and healthy immunocompetent PBMC stimulated to proliferate, while the cytotoxic actions exerted on unstimulated PBMC were less pronounced. The tested extracts exhibited considerably stronger cytotoxic activities towards HeLa, Fem-x and K562 cells in comparison to resting and stimulated PBMC. It is worth noting that the cytotoxicity of the extracts was weaker against unstimulated PBMC in comparison to stimulated PBMC. Furthermore, each of the five extracts induced apoptosis in HeLa cells, through the activation of both intrinsic and extrinsic signaling pathways. Conclusion: Extracts obtained from the endemic plant Helichrysum zivojinii may represent an important source of novel potential antitumor agents due to their pronounced and selective cytotoxic actions towards malignant cells.

AB  - Helichrysum zivojinii Cernjavski & Soska is an endemic plant species that grows in the National Park Galicica in Macedonia. Five extracts were isolated as fractions from the aerial parts of the plant: a n-hexane extract (1), a dichloromethane extract (2), an ethyl-acetate extract (3), a n-butanol extract (4) and a methanol extract (5). A dose-dependent cytotoxic activity of the extracts on MDA-MB-231 and EA.hy926 cells was observed. Extracts exhibited more pronounced cytotoxic actions on MDA-MB-231 cells than on EA.hy926 cells. The n-hexane extract (1), at a non-toxic concentration, exhibited an inhibitory effect on the migration as well the invasiveness of MDA-MB-231 cells. The dichloromethane extract (2), at a non-toxic concentration, demonstrated inhibition of MDA-MB-231 cells invasion. Each of the five extracts applied at non-toxic concentrations inhibited migration of EA.hy926 cells. The prominent inhibitory effect of the n-hexane extract on EA.hy926 cells migration was associated with a notable anti-angiogenic action of this extract. The other four tested extracts demonstrated mild anti-angiogenic activity. Our data highlight the prominent anticancer potential of n-hexane (1) and dichloromethane (2) extracts, which could be attributed to their very pronounced and selective cytotoxic activities as well as their anti-invasive and anti-angiogenic properties.

AB  - Multi-drug resistance (MDR) is a major obstacle to successful cancer treatment. Therefore, in vitro models are necessary for the investigation of the phenotypic changes provoked by cytotoxic agents and more importantly for preclinical testing of new anticancer drugs. We analyzed chromosomal, numerical, and structural changes after development of MDR, alterations in p53 and PTEN, single nucleotide polymorphisms (SNPs) in the mdr1 gene and corresponding protein expression of P-glycoprotein (P-gp) in three human MDR cancer cell lines: non-small cell lung carcinoma NCI-H460/R, colorectal carcinoma DLD1-TxR, and glioma U87-TxR. In addition, we explored how these molecular and phenotypic alterations influence the anticancer effect of new drugs. Cytogenetic analysis showed polyploidy reduction after development of MDR in U87-TxR. Losses of 6q in all resistant cancer cell lines and inactivation of p53 in U87-TxR and PTEN in DLD1-TxR were also revealed. Overexpression of P-gp was observed in all MDR cancer cell lines. We evaluated the anticancer activities and MDR reversal potential of Akt inhibitor GSK690693, Ras inhibitor Tipifarnib, and two P-gp inhibitors (jatrophane diterpenoids). Their effects vary due to the cell-type differences, existence of MDR phenotype, presence of mdr1 SNP, and tumor suppressors' alterations. Tipifarnib and jatrophane diterpenoids significantly sensitized MDR cancer cells to paclitaxel. In conclusion, investigated MDR cancer cells obtained new molecular and cytogenetic characteristics that may serve as potential clinical prognostic markers. In addition, these MDR cancer cell lines present a valuable model for preclinical evaluation of new anticancer agents.

AB  - Jatrophane diterpenes were shown to be inhibitors of P-glycoprotein (P-gp). There are also evidences on their microtubule-interacting activity in cancer cells. We evaluated new anti-cancer characteristics of two jatrophane type compounds from Euphorbia dendroides. For that purpose, the model system of sensitive non-small cell lung cancer cell line (NCI-H460) and its resistant counterpart (NCI-H460/R) was used. Although both jatrophanes showed inhibitory effect on cancer cell growth, they were non-toxic for peripheral blood mononuclear cells (PBMC). We examined their effects in combination with paclitaxel (PTX), a well-known mitotic spindle interacting chemotherapeutic. Jatrophanes overcome PTX resistance in concentration-dependent manner in MDR cancer cell line (NCI-H460/R). We observed that this synergistic effect is not caused merely by P-gp inhibition. In combination with PTX, jatrophanes induce cell killing and change cell cycle distribution leading to G2/M arrest. Furthermore, they exert an anti-angiogenic effect by decreasing the vascular endothelial growth factor (VEGF) secretion. The reduction of the level of mdr1 mRNA expression in sensitive cells, suggests that these compounds could not contribute to the development of resistance. In conclusion, present study provides a rational basis for the new cancer treatment approach with jatrophanes that are non-toxic to normal cells and have new favorable anti-cancer characteristics.

AB  - From the Montenegrin spurge Euphorbia dendroides, seven new diterpenoids [jatrophanes (1-6) and a tigliane (7)] were isolated and their structures elucidated by spectroscopic techniques. The biological activity of the new compounds was studied against four human cancer cell lines. The most effective jatrophane-type compound (2) and its structurally closely related derivative (1) were evaluated for their interactions with paclitaxel and doxorubicin using a multidrug-resistant cancer cell line. Both compounds exerted a strong reversal potential resulting from inhibition of P-glycoprotein transport.

Using the Cytochalasin-B blocked micronucleus (CBMN) assay, in which the biomarker of chromosome breakage and/or chromosome loss is the elevated frequency of micronucleus (MN) in binucleated (BN) cells, the presence of flavonoid 2 in minimal concentration (3 mu g/mL) gave a 35.5% decrease M the frequency of MN when compared with control human lymphocytes. The same concentration of flavonoids 1, 3 and 4, reduced the MN frequency by 24.4%, 28.0% and 28.0%, respectively. Higher concentrations (6 mu g/mL and 10 mu g/mL) seemed less effective. Flavonoid 5 (3 mu g/mL.) induced a slight decrease in MN frequency (5%), while higher doses (6 mu g/mL and 10 mu g/mL) provoked an increase of DNA damage. The comparable values for the cytokinesis-block proliferation index (CBPI) of the tested flavonoids and positive control suggested an inhibitory effect on lymphocyte proliferation. In the DPPH scavenging assay, flavonoids 1-4 demonstrated modest activity, in a dose-dependent manner, compared with the synthetic antioxidants BHT and Trolox, while 5 exhibited comparably high antioxidative activity.

AB  - The essential oil from aerial parts of Tanacetum larvatum (gris.) Kanitz, an endemic perennial herb collected at three various natural habitats in Montenegro (Planinica [Sample a ], Visitor [Sample b ] and Sinjajevina [Sample c ]), were analyzed using GC and GC/MS. Sixty-four components were identified, representing 83.1%, 96.6% and 89.4% of the total oils content in the Samples a, b and c , respectively. The major constituent in Samples a and b , was oxygenated monoterpene, trans-sabinyl acetate (38.1% and 55.8% respectively). Monoterpene hydrocarbons, β-pinene (13.5%) and santolinatriene (30.6%), were found to be the dominant components in Sample c . The toxic trans-sabinyl acetate was present only in traces in this sample. trans-Chrysanthenyl acetate, as one of major components in feverfew essential oil, has not been previously identified in the investigated essential oils.

AB  - Tanacetum larvatum (Oris.) Kanitz. is an endemic perennial herb distributed on a rocky terrain in Serbia (Kosovo), Montenegro and Albania. The chemical composition of T. larvatum essential oil, obtained from two locations in Montenegro (Mt. Komovi [Sample I] and Mt. Prokletije [Sample II]) was investigated. The oils obtained by hydrodistillation were analyzed by GC and GC/MS, on the two different capillary columns. The aerial parts of T. larvatum yielded 0.3% and 0.2% of a yellowish oil (Sample I and Sample II, respectively). About 40 compounds were identified, representing ∼89% and 96% of the total oil content in the Samples I and II, respectively. trans-Sabinyl acetate was found to be the dominant component (51.2% and 69.7%). Among the rest of compounds β-pinene (7.7% and 4.3%) and camphor (6.3% and 4.3%) were the most abundant in both samples.

AB  - As shown by quantitative 1H NMR measurements, a lipophilic extract of the aerial parts of Hypericum atomarium ssp. degenii contained a high percentage (3.1% per weight of dried plant material) of a prenylated phloroglucinol (1). Compound 1, named hyperatomarin, occurring in two tautomeric forms (1a ⇆ 1b), was isolated by bioactivity-guided preparative TLC and was identified on the basis of spectral data interpretation. This isolated phloroglucinol exhibited activity against Gram-positive (Staphyloccocus aureus and Microccocus luteus) and Gram-positive spore-forming bacteria (Bacillus subtilis and B. IP 5832).

AB  - The conformations of the C-6 lactonized germacranolides 1–3 were calculated by applying the PM3 semi-empirical method. The low-temperature 1H-NMR spectra of parthenolide (1) were also measured. The relations between the calculated and the experimentally determined geometries were established using a modified Karplus equation. The applied quantum-chemical calculations proved to be an efficient and easy-to-use tool for the elucidation and prediction of the properties of germacranolide-type sesquiterpene lactones.
AB  - Konformacije germakranolida 1–3, laktonizovanih u položaju (6), izračunate su primenom PM3 semi-empirijske MO metode. Takođe su snimljeni protonski NMR spektri partenolida (1) na niskim temperaturama. Pomoću modifikovane Karplusove jednačine je uspostavljena korelacija između eksperimentalno utvrđenih i izračunatih najstabilnijih konformacija. Zaključeno je da primenjena kvantno-hemijska izračunavanja brzo i pouzdano mogu predvideti i objasniti osobine seskviterpenskih laktona tipa germakranolida.

The low-temperature 1H-NMR spectra of parthenolide (1) were also measured. AB  - Isolation of 1-O-primeverosyl-3,8-dihydroxy-5-methoxyxanthone and 1-O-gentiobiosyl- 3,7-dimethoxy-8-hydroxyxanthone, along with five known xanthones, isobellidifolin, methylbellidifolin, isoswertianin, methylswertianin and norswertianin-1-O-beta-D-glucoside, from the roots of Swertia punctata is reported. In the aerial parts four xanthones, bellidifolin, methylbellidifolin, swertianolin and mangiferin, and flavone-C-gluco side, isoorientin were identified. AB  - The genus Tanacetum L. comprises ca 150 species that commonly occur in Europe from the Mediterranean to Persia. Tanacetum larvatum (Gris.) Kanitz, the examination of which is reported, is distributed on elevated rocky terrains in Albania, Serbia (Kosovo) and Montenegro. Aerial parts of T. larvatum were collected in Montenegro on two locations: crop a on mountain Komovi (altitutude 1800 m, 12 July 1998) and crop b on mountain Babji Zub (altitude ca 1500 m, 30 June 1999).

AB  - Four flavones (1-4) and nine sesquiterpene lactones (5-13), one of them (5) a new compound, were isolated from the aerial parts of Achillea atrata L. subsp, multifida. Although the crude extract demonstrated in vitro inhibitory activity against Candida albicans and Bacillus subtilis, all isolated flavones were active against B. subtilis. Flavones 1, 2, and 3 were also active against C. albicans, while 1 and 3 exhibited activity against E. coli, as well. None of the tested lactones (7, 9, 12, and 13) showed any antimicrobial activity.

AB  - The isolation of three highly oxygenated guaianolides, desacetyl-1α-4α- dihydroxybishopsolicepolide [1], its 4β-hydroxy epimer 2 (a new compound), and 1β,2β:3β,4β-diepoxy-8α,10α-dihydroxyguai-12(13)-en-12,6α-olide [3], from the aerial parts of Achillea crithmifolia is reported. Because lactone 3 was shown to be identical with rupin A, the initially proposed 1α,2α:3α,4α-diepoxy stereochemistry of rupin A should be revised.
