dc.description.abstract | Četiri izoforme eksterne invertaze (EINV1, EINV2, EINV3 i EINV4) iz kvasca S. cerevisiae izolovane su izoelektričnim i etanolnim taloženjem, jonoizmenjivačkom hromatografijom na različitim QAE kolonama i gel hromatografijom. Za razliku od prethodno publikovanih procedura za izolovanje eksterne invertaze, primenom specijalno dizajniranog stepenastog gradijenta elucije sa jonoizmenjivačke kolone omogućeno je razdvajanje izoformi. Ovime je pokazano da je ono što je prethodno smatrano eksternom invertazom u stvari smeša bar četiri izoforme. Izoforme imaju identične katalitičke osobine, pH optimum i temperaturni optimum. MALDI-TOF masenom spektrometrijom određena molekulska masa izoformi EINV1, EINV3 i EINV4 je 104 kDa, dok izoforma EINV2 ima ~ 3 kDa manju masu. 31P-NMR spektroskopijom pokazano je da izoforme sadrže dva tipa fosfata vezanog za oligomanoznu komponentu. Najveći deo, 95 % fosfata nalazi se diestarski vezan za dve manozne jedinice, a ostatak je vezan za manozu u obliku monofosfata. Izoforme se značajno razlikuju u pI vrednosti, pokretljivosti u nedenaturišućoj elektroforezi, termalnoj stabilnosti, stabilnosti u metanolu i hemijskoj reaktivnosti. Sve ove razlike u korelaciji su sa gustinom negativnog naelektrisanja na površini izoformi koja raste redom kojim se izoforme eluiraju sa jonoizmenjivačke kolone (tj. od EINV1 do EINV4). Enzimskim i hemijskim deglikozilovanjem pokazano je da razlike između izoformi potiču od posttranslacionih modifikacija. Različita količina ukupnih fosfata vezanih za oligomanoznu komponentu izoformi ne može da objasni razliku u gustini površinskog naelektrisanja između izoformi EINV1 i EINV2 zato što izoforma EINV2 sadrži najmanju količinu ukupnog fosfata. CD i fluorescentnom spektrometrijom pokazano je da izoforme imaju slične strukturne osobine. Termodinamički parametri razvijanja izoformi (ΔGo, ΔHo, TΔSo, ΔCpo) izračunati su globalnom termodinamičkom analizom kriva denaturacije gvanidinijum-hloridom, koje su opisane reverzibilnim modelom dva stanja. Izoforme pokazuju različite termodinamičke parametre, a razlike su u direktnoj korelaciji sa gustinom negativnog naelektrisanja na površini izoformi. Pored teorijskog značaja za razumevanje uticaja gliko-komponente na biofizičke osobine glikoproteina ova istraživanja imaju i praktičnu primenu. Diskutovana je potencijalna primena izoforme EINV1, u cilju povećanja efikasnosti industrijskih procesa u kojima se koristi invertaza,s obzirom na to da pokazuje najveću termalnu i stabilnost u metanolu, kao i značajno veću hemijsku reaktivnost prema eupergitu C. | sr |
dc.description.abstract | Four isoforms (EINV1, EINV2, EINV3 and EINV4) of yeast S. cerevisiae external invertase
were purified by isoelectric and ethanol precipitation, ion-exchange chromatography on QAE
columns and gel filtration. Unlike previously published procedures for external invertase
purification, specially designed step elution was applied on ion-exchange column which
enabled separation of isoforms. Results showed that what has been previously considered for
external invertase is a mixture of at least four isoforms. The isoforms have the same catalytic
properties, pH optimum and temperature optimum. Molecular mass of isoforms determined
by MALDI-TOF mass spectrometry is 104 kDa for EINV1, EINV3 and EINV4, while for
EINV2 is ~ 3 kDa lower. 31P-NMR spectroscopy measurements showed that isoforms
contains two types of phosphate bound for oligomannose component. Biggest part, 95 % of
phosphate is in phosphodiester bond between two mannose units. Remaining phosphate is
bound for mannose as monophosphate. Isoforms exhibit significant difference in pI values,
thermal stability, stability in methanol and chemical reactivity toward eupergit C. These
differences are in correlation with negative charge density at the surface of isoforms.
Negative charge density of isoforms increases in the same order as they are eluted from ionexchange columns (i.e. form EINV1 to EINV4). Enzyme and chemical deglycosylation
studies showed that the observed differences between isoforms arise from posttranslational
modifications. Different amount of the total phosphate bound for oligomannose component
cannot explain the difference in the surface negative charge density between EINV1 and
EINV2 since EINV2 contains lower amount of bound phosphate. CD and fluorescence
spectroscopy showed that isoforms share similar structural features. Thermodynamic

) of unfolding of isoforms were calculated by global
thermodynamic analysis of guanidinium chloride induced unfolding, which has been
described by reversible two-state model. Isoforms exhibit different thermodynamic
parameters and they are in correlation with negative surface charge density. Beside
theoretical significance for understanding influence of glyco component to biophysical
properties of glycoproteins, this research has a practical application. Potential application of
isoform EINV1 has been discussed in order to improve efficiency of industrial processes
involving invertase, since EINV1 exhibit highest thermal and stability in methanol and higher
