AB  - Chokeberries (Aronia melanocarpa (Michx.) Elliot have recently become one of the most popular and widely used berry fruits, and there are numerous studies dealing with the investigation of their chemical composition and biological activities. Unlike berries, chokeberry leaves were far less investigated. Therefore, the aim of this study was to conduct a bioactivity-based analysis of chokeberry leaves collected after berries were harvested. Chemical analysis of hydroethanolic chokeberry extract and its ethyl acetate, n-butanol, and water fractions was performed. Antioxidant, hypoglycemic, and antineurodegenerative activities of the obtained samples were assessed by in vitro tests. The highest total phenolics content was found in n-butanol fraction (221.5 mg gallic acid equivalents/g dw). Fourteen compounds (six phenolic acids and eight flavonoids) were identified in the samples. Caffeoylquinic acid derivatives and quercetin glycosides were dominant phenolic compounds. The highest content of phenolic acids, as well as flavonoids, was recorded in n-butanol fraction (80.75 mg/g dw and 47.79 mg/g dw, respectively). This fraction was the most active in 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis (3-ethyl-benzothiazoline-6-sulfonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) assays. Also, the n-butanol fraction was the most effective inhibitor of acetylcholinesterase (AChE) and tyrosinase (TYR) enzymes. All samples showed a better inhibitory effect on α-glucosidase enzyme with inhibition concentrations IC50 = 4.21 - 9.22 μg/mL, than α-amylase enzymes (IC50 = 14.36 - 88.61 μg/mL). Pearson’s correlation between the content of individual phenolics and the results of biological tests was performed. Thus, hyperoside and isoquercitrin showed a significant correlation with DPPH and ABTS tests, caffeoylquinic acids with FRAP test, quercetin 3-O-vicianoside with β-carotene assay, hyperoside and isoquercitrin with α-amylase inhibition activity, and isorhamnetin 3-O-rutinoside with α-glucosidase inhibition assay. Considering the obtained results, chokeberry leaves represent a valuable natural source of phenolic compounds with promising potential for the development of new products with beneficial effects on human health.

Grape Seed Polyphenols and Fatty Acids of Autochthonous Prokupac Vine Variety from Serbia

AB  - Grape pomace (grape skin, seeds, and stems) represents a valuable source of bioactive compounds, thus the interest in making use of these wine waste products is increasing. Prokupac is an autochthonous Serbian variety of black grape with a long tradition in production of red wine. Identification of the superficial polyphenolic compounds by LC/MS revealed presence of 19 compounds including hydroxybenzoic acid derivatives, ellagic acid derivatives, flavan‐3‐ol monomers, proanthocyanidins, and flavonols. Catechin and epicatechin together with proanthocyanidins were the most abundant compounds and the highest content of phenolic compounds was determined in Prokupac clones 43/1, 40/1, and 40/2. Grape seed oil consisted of dominantly UFA (87–95 %) where methyl linoleate was the most abundant in all samples (69–81 %). Palmitic (3–8 %) and stearic acid (2–4 %) methyl esters were dominant SFA. Prokupac clone 43/3 was the most abundant in UFA (95 %) with the lowest percentage of SFA (5 %), while clones 41/1 and 43/5 had the lowest percentage of UFA (87 %) and the highest content of SFA (13 %). Favorable chemical profile of Prokupac clone seeds implicates its potential to be used as a raw material in further processing and possibility to be included as one of the quality parameters in further selection of the most interesting Prokupac clone.

AB  - Despite the fact that Thymus serpyllum is well-known medicinal plant and its chemical profile and biological activity have been investigated, there is no detailed study regarding the influence of different techniques and conditions on the extraction of polyphenolic compounds from Serpylli herba. The aim of this study was optimization of the extraction parameters that improves the efficiency of polyphenols extraction from T. serpyllum: particle size, solid-to-solvent ratio, solvent type and extraction time, by using maceration, heat- and ultrasound-assisted extraction (HAE and UAE). The extraction efficiency was expressed via total polyphenol content (TPC) and total flavonoid content (TFC). The statistical analysis (one-way ANOVA and full factorial design) has revealed that the optimal conditions for achieving the best polyphenols yield were particle size of 0.3 mm, 1:30 solid-to-solvent ratio and 50% ethanol, as environmentally friendly extraction medium, while extraction time has not shown statistically significant influence on polyphenols concentration, in all procedures. Under these conditions, the measured TPC was 26.6 mg GAE/L in maceration, 29.8 mg GAE/L in HAE and 32.7 mg GAE/L in UAE, which was in agreement with the predicted values, while TFC was 143 mg CE/L, 12.4 mg CE/L and 16.7 mg CE/L for maceration, HAE and UAE, respectively. According to total polyphenols yield, the efficiency of the extraction methods for all variables was ranked by significance in the following order: UAE > HAE > maceration, whereas total flavonoids yield was the highest in UAE, although there was no statistically significant difference between maceration and HAE. According to our results, UAE could be selected as the most successful and suitable technique for extraction of bioactive polyphenolic compounds from Serpylli herba. Using LC/MS and HPLC analysis, 9 polyphenolic compounds were identified and quantified: 6,8-Di-C-glucosylapigenin, chlorogenic acid, 6-hydroxyluteolin 7-O-glucoside, caffeic acid, luteolin 7-O-glucuronide, apigenin glucuronide, salvianolic acid g isomer, rosmarinic acid and salvianolic acid I. This study was an initial step in production of polyphenols-rich wild thyme extracts aimed to be used for formulation of foodstuffs and medicines.

According to our results, UAE could be selected as the most successful and suitable technique for extraction of bioactive polyphenolic compounds from Serpylli herba. Using LC/MS and HPLC analysis, 9 polyphenolic compounds were identified and quantified: 6,8-Di-C-glucosylapigenin, chlorogenic acid, 6-hydroxyluteolin 7-O-glucoside, caffeic acid, luteolin 7-O-glucuronide, apigenin glucuronide, salvianolic acid g isomer, rosmarinic acid and salvianolic acid I. This study was an initial step in production of polyphenols-rich wild thyme extracts aimed to be used for formulation of foodstuffs and medicines.",

AB  - The aim of this study was the determination of phenolic compounds, and mineral nutrients in the pulp and peel obtained from Balkan indigenous apple and pear cultivars. The phenolic composition, assessed by HPLC-DAD and spectrophotometric methods varied significantly between the cultivars for both peel and pulp. Among the apples, the Mekica cultivar had the highest total phenolic content while Secerlija and Zlatna Parmenka had the lowest. In the case of the pear samples, the maximum total phenolic content was found in Crna Takisa while the minimum content was recorded for Bela Arapka. In most of the investigated samples, chlorogenic acid in apples and arbutin in pears were the major detected polyphenolic compounds. With regard to the mineral analysis, K was the most abundant followed by P, Mg, Ca and S. Iron was the dominant microelement in apple peel and pulp samples, while in pear samples the principal microelement was B. The obtained results provide detailed information on the chemical composition of the tested apple and pear cultivars and thereby, could encourage their wider cultivation and consumption.

AB  - Pumpkin fruit is used in a diet since ancient times especially in rural communities. The major contributory factors of nutritional and medicinal value of pumpkins are carotenoids, polysaccharides, vitamins, minerals, and phenolic compounds. Due to a very large fruit that it is not easy to consume a whole as well as short shelf-life of fresh-cut pumpkin, different ways of conserving and processing are performed. In our study, total carotenoids, total phenolics and individual phenolics in fresh pumpkin and pumpkin traditional products such as sweet in wine, jam and juice, which are typical for northern parts of Serbia, were studied. Total carotenoids ranged from 27.6 mu g/g of pumpkin sweet in wine to 86.3 mu g/g of fresh fruit, while the amount of total phenolics varied between 93.0 mu g GAE/g of pumpkin juice and 905.9 mu g GAE/g of fresh fruit. Eight phenolic compounds were identified in the investigated samples and among them phenolic acids dominated. Among flavonoids, flavanon glycoside hesperidin was detected.

AB  - Wine quality depends mainly on the characteristics of the grape it is made of, and one of the attributes affecting wine composition is cluster and berry morphology. The aim of this study was to represent variability of the morphological characteristics between different clones of the autochthonous grape variety Prokupac and to perform chemical evaluation of wines obtained from them. Total phenolic content was generally low and it ranged from 33.0 to 114.5 mg GAE/100 mL. Six main anthocyanin compounds including malvidin as the main anthocyanidin were detected. Malvidin 3-O-glucoside was the most abundant anthocyanin with concentration of 59.8 to 101.7 mu g/mL. Clones 43/5 and 43/4 yielded highest quality wines. According to the results, clonal selection makes a significant difference in Prokupac wine quality. On the other hand, there is a minor dependance of wine quality parameters to variation in morphological attributes of clusters and berries (bunch weight, proportion of stem, berry and seed weight, skin, pulp and seed weight per berry).

Optimization of extraction of stinging nettle leaf phenolic compounds using response surface methodology

AB  - A full three level factorial design was implemented for optimization of extraction parameters in order to maximize total phenolic (TP) yield from stinging nettle leaf. Factors considered were percentage of methanol in solvent (X-1 : 50-100% methanol) and extraction time (X-2: 30-90 min), and maceration was used as extraction method. A second-order polynomial model was applied for fitting experimental data and predicting the response, and this mathematical model produced a satisfactory fit (R-2 = 0.993,p LT 0.01). The optimal extraction conditions were 54% aqueous methanol and 38 min extraction time, while maximal theoretical TP yield was 8.9 mg GAE/g DW. Solvent composition significantly affected extraction efficiency causing decrease of TP yield with increase of methanol percentage. On the other hand, extraction time did not influence significantly efficiency of extraction. Using LC/MS and HPLC analysis we detected and quantified three most abundant phenolic compounds: 2-O-caffeoyl malic acid, chlorogenic acid, and rutin. Comparison between maceration and ultrasound-assisted extraction (UAE) obtained extracts based on TP content as well as antiradical activity and HPLC results, showed that UAE have a better extraction capability affecting yield and time of extraction. Of all tested extracts, 54% aqueous methanolic extracts obtained with UAE and 38 min extraction time had the highest TP content.

AB  - A detailed phenolic composition analysis of dry chokeberry, bilberry and black currant fruit teas prepared on the most common ways-decoction and infusion, was performed, together with evaluation of cytotoxic activity. The most abundant in phenolics were chokeberry teas, followed by bilberry and black currant teas, while the highest anthocyanin amount was determined in bilberry samples. LC/DAD/MS method was used for identification of 17 anthocyanins, 11 flavonoids and 4 hydroxycinnamic acid derivatives. Quantitative analysis of investigated teas was carried out by HPLC analysis, and major phenolic compounds in berry fruit teas were chlorogenic acids, quercetin glycosides and anthocyanin glycosides. Berry teas were investigated for cytotoxic activity against cervix carcinoma, melanoma, colon, and chronic myelogenous leukemia cell lines, and chokebeny decoction was the most effective against all cell lines. According to the results obtained in this study, pure chokeberry, bilberry and black currant teas could be valuable sources of flavonoids and anthocyanins.

AB  - In recent years great attention was paid to biological control, the application of natural products in order to protect crops and medicinal plants against contamination with phytopathogenic and saprophytic fungi. Essential oils have been evaluated as a potentially safe replacement for chemicals used for that purpose. In this regard, antifungal potential of six essential oils was examined coupled with determination of the complete qualitative and quantitative chemical composition by GC-FID and GC-MS analysis. While essential oils of rosemary, sage, Spanish sage and black pepper were exclusively composed of monoterpenes, vetiver essential oil was entirely composed of sesquiterpenes. In essential oil of cinnamon dominates (E)-cinnamaldehyde. Applying the in vitro microdilution method, it was found that all essential oils were active in inhibiting the growth of all tested 21 pre- and post-harvest phytopathogenic and saprophytic fungi. MIC and MFC ranged from 1.2 mg ml-1 up to 22.6 mg ml-1 according to the test oil. Rosemary oil showed the best antifungal potential, followed by black pepper and cinnamon oil. Sage and Spanish sage oils also exhibited significant antifungal potential. Vetiver oil demonstrated the lowest antifungal activity. Essential oils that showed considerable antifungal potential are good candidates for further examination of their use in preventing and/or protection of medicinal plants, their seeds and dried drugs against fungal infections, both in the field and in warehouses.
AB  - Poslednjih godina velika pažnja poklanja se biološkoj kontroli to jest primeni prirodnih produkata u zaštiti, kako poljoprivrednih kultura, tako i lekovitog bilja od kontaminacije fitopatogenim i saprofitnim gljivama. U tom smislu, ispitivan je antifungalni potencijal šest etarskih ulja uz određivanje kompletnog kvalitativnog i kvantitativnog hemijskog sastava primenom GC-FID i GC-MS analiza. Dok se korišćeni uzorci etarskih ulja ruzmarina, žalfije, španske žalfije i crnog bibera isključivo sastoje od monoterpena, u ulju vetivera potpuno dominiraju seskviterpeni. U etarskom ulju cimeta najzastupljenija komponneta je bila (E)- cinemaldehid. Primenom in vitro mikrodilucione metode, utvrđeno je da su sva ulja bila aktivna u inhibiciji rasta 21 testirane, pre- i post-žetvene, fitopatogene i saprofitne gljive, sa razlikama u efikasnosti. Etarska ulja koja poseduju značajan antifungalni potencijal su dobri kandidati za nastavak istraživanja u pravcu njihove primene u prevenciji i zaštiti useva lekovitog bilja, kao i njihovih semena i osušenih biljnih droga od gljivičnih infekcija, i u polju i u skladištima.

Phenolic and mineral profiles of four Balkan indigenous apple cultivars monitored at two different maturity stages

AB  - Some primary and secondary metabolites, as well as mineral nutrients in pulp, peel and juice obtained from four Serbian indigenous apple cultivars (Kozara, Kolacara, Budimka and.Sumatovka) collected at two different developmental stages were studied. With advanced maturation soluble solids content, total and reducing sugars increased, while L-ascorbic acid content and titratable acidity decreased. Thirteen phenolic compounds were quantified using LC-MS/MS. The total phenolic content (TPC) ranged from 9.37 to 1440 mg/100 g fw, and 0.83 to 7.84 mg/100 g fw in peel and pulp samples, respectively. Quercetin derivatives were the major detected polyphenolic group. Majority of determined phenolic compounds were influenced by cultivar and the best sources were cultivars Kolgara and Budimka. The content of flavonols (with the exception of quercitrin) varied significantly depending on maturity at harvest. With regard to mineral analysis, K was the most abundant ranging from 104 to 158 mg/100 g fw in peel, 74.4 to 93.3 mg/100 g fw in pulp, and 77.1 to 91.5 mg/100 g fw in juice samples. Obtained results provide detailed information on nutritional potential and chemical composition of tested apple cultivars and thereby could encourage their wider cultivation and consumption

AB  - Phenolic compounds are very important quality parameters of wine because of their impact on colour, taste and health properties. The present study was aimed at evaluating the general phenolic composition and free radical scavenging activity of aqueous and organic fractions obtained using liquid liquid extraction of red wine produced from the Serbian autochthonous grape variety Prokupac. The total phenolic contents in the different fractions ranged from 48.22 to 289.12 mg GAE per g dry fraction. Phenolic acids (mainly hydroxycinnamic acids) and quercetin 3-O-glucuronide were the main components in the EtOAc fraction at pH 2.0; catechins, phenolic acids (mainly hydroxybenzoic acids) and quercetin were found in the EtOAc fraction at pH 7.0, while anthocyanins were identified in the aqueous residue after EtOAc extraction. The major anthocyanin extracted into the aqueous fraction was malvidin-3-glucoside, while the most abundant non-anthocyanin phenolic compounds in the organic fractions were ethyl gallate and trans-caftaric acid. The radical scavenging activities of the fraction differed significantly and the IC50 values were 138.58 mu g mL(-1) for the aqueous fraction, 17.83 and 3.47 ng mL(-1) for the EtOAc fractions at pH 2.0 and 7.0, respectively. As the EtOAc fractions were found to be more potent radical scavengers, it could be assumed that non-anthocyanin phenolic compounds were responsible for such activity in Prokupac wine.

Qualitative and quantitative chemical analysis of 16 selected essential oils was determined. Then, antifungal properties determined by in vitro microdilution method against 21 fungi isolated from herbal drugs were evaluated. All tested oils showed some antifungal activity against all fungi used. Savory, thyme and oregano oils, characterized by the presence of phenol such as carvacrol and thymol, and rose oil containing mainly monoterpene alcohols (citronellol and geraniol) proved to be the most effective inhibitor of all fungi tested. Also, combination of particular oils showed reduction of the MIC values when combined, commendatory mixtures for potential application in practice. Moreover, the reduction of the total number of fungi, in situ, using selected essential oils was determined.

Morphological and chemical characterization of Arnica montana L. under different cultivation models

AB  - Arnica (Arnica montana L) is a perennial mountain plant whose flower heads are mainly used for topical treatment of bruises and sprains in phytopharmaceutical preparations such as ointments, creams and gels. In this study, different models of arnica cultivation have been tested in three-year field trial arranged in 2 x 3 x 2 factorial design. Planting date (spring and autumn), fertilizater type (manure, NPK and control) and mode of reproduction (generative and clonal) were induced factors on the trial. The variation of the rosette diameter, flowering stem count and height, flower heads count and diameter, together with flower heads yield and chemical properties were investigated as morphological parameters. Spring planting of clonally propagated plants was showed to be the least successful model due to the low percent of rooted plants and weak shoot development. Other models yielded from 59.8 to 143.6 kg ha(-1) and 116.3 to 258.7 kg ha(-1) flower heads (dw) in the second and third year, respectively. Total sesquiterpene lactones content in flower heads of all tested variants was above the minimum of pharmacopeia quality regulation and ranged from 4.6 to 13.9 mg g(-1) depending on factor levels, while the amounts of two dominant flavonoid compounds quercetin-3-O-glucoside and kaempferol-3-O-glucoside ranged from 7.8 to 13.9 mg g(-1) and from 2.1 to 4.7 mg g(-1), respectively. All induced factors in the field trial significantly influenced variation in both morphological and chemical parameters, where fertilized variants from autumn planting have shown to be superior in flower heads yield with satisfying levels of the secondary metabolites content. Factorial analysis distinguished that half of morphological parameters positively correlated with flower heads yield. First three ranks of sequential classification of cultivation models with desirable properties were captured by variants planted in autumn, while the last rated variants were those from clonal propagation planted in spring. This attempt of arnica domestication in agroecological conditions of western Serbia has proven to be successful in ensuring high quality raw material, providing at the same time answers to the questions that have not been reviewed so far.

abstract = "Arnica (Arnica montana L) is a perennial mountain plant whose flower heads are mainly used for topical treatment of bruises and sprains in phytopharmaceutical preparations such as ointments, creams and gels. In this study, different models of arnica cultivation have been tested in three-year field trial arranged in 2 x 3 x 2 factorial design. Planting date (spring and autumn), fertilizater type (manure, NPK and control) and mode of reproduction (generative and clonal) were induced factors on the trial. The variation of the rosette diameter, flowering stem count and height, flower heads count and diameter, together with flower heads yield and chemical properties were investigated as morphological parameters. Spring planting of clonally propagated plants was showed to be the least successful model due to the low percent of rooted plants and weak shoot development. Other models yielded from 59.8 to 143.6 kg ha(-1) and 116.3 to 258.7 kg ha(-1) flower heads (dw) in the second and third year, respectively. Total sesquiterpene lactones content in flower heads of all tested variants was above the minimum of pharmacopeia quality regulation and ranged from 4.6 to 13.9 mg g(-1) depending on factor levels, while the amounts of two dominant flavonoid compounds quercetin-3-O-glucoside and kaempferol-3-O-glucoside ranged from 7.8 to 13.9 mg g(-1) and from 2.1 to 4.7 mg g(-1), respectively. All induced factors in the field trial significantly influenced variation in both morphological and chemical parameters, where fertilized variants from autumn planting have shown to be superior in flower heads yield with satisfying levels of the secondary metabolites content. Factorial analysis distinguished that half of morphological parameters positively correlated with flower heads yield. First three ranks of sequential classification of cultivation models with desirable properties were captured by variants planted in autumn, while the last rated variants were those from clonal propagation planted in spring. This attempt of arnica domestication in agroecological conditions of western Serbia has proven to be successful in ensuring high quality raw material, providing at the same time answers to the questions that have not been reviewed so far.",

Chamomile and Marigold Tea: Chemical Characterization and Evaluation of Anticancer Activity

AB  - With the aim to evaluate the selectivity in the antitumor action, the cytotoxic activity of chamomile and marigold tea was tested against various malignant cell lines and against healthy immunocompetent peripheral blood mononuclear cells (PBMC). Chemical profiles of chamomile and marigold infusions and decoctions were analyzed by liquid chromatography/mass spectrometry; their total phenolic content and radical scavenging activity were determined, too. Results from present research demonstrate that chamomile and marigold tea exert selective dose-dependent cytotoxic action against target cancer cells. It is noteworthy that cytotoxicity of tea prepared from Calendula officinalis is remarkably higher in comparison to that from Matricaria recutita tea. The cytotoxic effect of chamomile tea is very weak to healthy PBMC, while the effect of marigold tea on PBMC is more pronounced. Marigold tea exerts highly selective antitumor effect especially to melanoma Fem-x cells in comparison to the action to normal healthy PBMC. Chemical analyses show that dominant phenolic compounds in examined infusions and decoctions are flavonoid glycosides and hydroxycinnamic acid derivatives. There are no considerable differences in total phenolic content and antioxidant activity between examined infusions. Antitumor potential of chamomile and marigold tea should be further investigated.

AB  - Collection of Sideritis raeseri subsp. raeseri has a long tradition in local communities in the Ohrid-Prespa region. The aim of the present study was the analysis of bioactive compounds especially those with anti-inflammatory activity. Combination of the UV and MS data allowed the characterization of 17 compounds, which could be classified into flavonoid glycosides or hydroxycynnamic acid derivatives. Six of them were isolated using preparative HPLC: isoscutellarein 7-O-[6'''-O-acetyl-beta-D-allopyranosyl-(1 -> 2)]-beta-D-glucopyranoside, 4'-O-methylhypolaetin 7-O-[6'''-O-acetyl-beta-D-allopyranosyl-(1 -> 2)]-beta-D-glucopyranoside, hypolaetin 7-O-[6'''-O-acetyl-beta-D-allopyranosyl-(1 -> 2)]-6 ''-O-acetyl-beta-D-glucopyranoside, 4'-O-methylisoscutellarein 7-O-[6'''-O-acetyl-beta-D-allopyranosyl-(1 -> 2)]-beta-D-glucopyranoside, isoscutellarein 7-O-[6'''-O-acetyl-beta-D-allopyranosyl-(1 -> 2)]-6 ''-O-acetyl-beta-D-glucopyranoside and 4'-O-methylhypolaetin 7-O-[6'''-O-acetyl-beta-D-allopyranosyl-(1 -> 2)]-6 ''-O-acetyl-beta-D-glucopyranoside. AB  - LC-ESI-MS and HPLC were used for the identification of the constituents from G. lutea leaves collected at different localities, as well as for quantification of the main compounds. Seven secoiridoids, five C-glucoflavones and three xanthones, were identified. Swertiamarin derivatives, namely eustornorusside (2), eustomoside (3) and septemfidoside (5), were detected in G. lutea for the first time. Concentrations of five constituents (swertiamarin, gentiopicrin, isovitexin, mangiferin and isogentisin) were determined. The relationship between concentrations of gamma-pyrones and altitude was observed with statistically significant correlation (r = 0.94). The extracts were also evaluated for their content of total phenolics, and antiradical and cytotoxic activities. The total phenolics content ranged from 7.7 to 12.7 mg GAE/g, and the IC50 values for DPPH radical scavenging activity varied between 0.45 to 2.02 mg/mL. The leaf extract exhibited moderate cytotoxic effects toward HeLa cells with an IC50 value of 41.1 mu g/mL, while gentiopicrin, mangiferin and isogentisin exerted strong activity against HeLa cells, with IC50 values ranging from 5.7 to 8.8 mu g/mL. The results confirm the traditional usage of G. lutea leaves and also suggest their possible utilization as hepatoprotective, hypoglycemic and anti-inflammatory agents.
