AB  - Stress is a physiological reaction of an organism to a demand for change that is imposed by external factors or is coming from within by way of physiological strains or self-perceived mental and/or emotional threats (internal factors). It manifests itself through the sudden release of a flood of hormones, including corticosteroids, into the blood, which rouse the body for action. Normally, stress is beneficial, but when lasting or being very strong, it causes major damage to our mind and body. Despite intense research, we still do not understand fully how the stress response axis, whose main function is to respond to challenges while maintaining the normal physiological balance, loses under prolonged exposure to stressors its capacity to maintain homeostasis. Recent applications of mathematical modelling and dynamical systems theory have enabled us to emulate complex neurochemical transformations that underlie the stress response, and help us to acquire deeper understanding of this dynamical regulatory network.

Natural sorbents modified by divalent Cu2+- and Zn2+- ions and their corresponding antimicrobial activity

AB  - The objective of this study was to investigate the modification of materials used in wastewater treatment for possible antimicrobial application(s). Granulated activated carbon (GAC) and natural clinoptilolite (CLI) were activated using Cu2+-and Zn2+-ions and the disinfection ability of the resulting materials was tested. Studies of the sorption and desorption kinetics were performed in order to determine and clarify the antimicrobial activity of the metal-activated sorbents. The exact sorption capacities of the selected sorbents, GAC and CLI, activated through use of Cu2+-ions, were 15.90 and 3.60 mg/g, respectively, while for the materials activated by Zn2+-ions, the corresponding capacities were 14.00 and 4.72 mg/g,. The desorption rates were 2 and 3 orders of magnitude lower than their sorption efficacy for the Cu2+-, and Zn2+-activated sorbents, respectively. The intermediate sorption capacity and low desorption rate indicated that the overall antimicrobial activity of the metal-modified sorbents was a result of metal ions immobilized onto surface sites. The effect of antimicrobial activity of free ions desorbed from the metal-activated surface may thus be disregarded. The antimicrobial activities of Cu/GAC, Zn/GAC, Cu/CLI and Zn/ CLI were also tested against Escherichia coli, Staphylococcus aureus, and Candida albicans. After 15 min exposure, the highest levels of cell inactivation were obtained through the Cu/CLI and the Cu/GAC against E. coli, 100.0 and 98.24%, respectively. However, for S. aureus and yeast cell inactivation, all Cu2+-and Zn2+-activated sorbents proved to be unsatisfactory. A characterization of the sorbents was performed by Xray diffraction (XRD), X-ray photo electron spectroscopy (XPS), and field emission scanning electron microscopy (FE-SEM). A concentration of the adsorbed and released ions was determined by inductively coupled plasma-optical emission spectroscopy (ICP-OES) and mass spectrometry (ICP-MS). The results showed that the antimicrobial performance of the activated sorbents depended on the surface characteristics of the material, which itself designates the distribution and the bioavailability of the activating agent.

AB  - A model was developed that can be used to study the effect of gradual cholesterol intake by food on the HPA axis dynamics. Namely, well defined oscillatory dynamics of vital neuroendocrine hypothalamic-pituitary-adrenal (HPA) axis has proven to be necessary for maintaining regular basal physiology and formulating appropriate stress response to various types of perturbations. Cholesterol, as a precursor of all steroid HPA axis hormones, can alter the dynamics of HPA axis. To analyse its particular influence on the HPA axis dynamics we used stoichiometric model of HPA axis activity, and simulate cholesterol perturbations in the form of finite duration pulses, with asymmetrically distributed concentration profile. Our numerical simulations showed that there is a complex, nonlinear dependence between the HPA axis responsiveness and different forms of applied cholesterol concentration pulses, indicating the significance of kinetic modelling, and dynamical systems theory for the understanding of large-scale self-regulatory, and homeostatic processes within this neuroendocrine system.

Dynamic transitions in a model of the hypothalamic-pituitary-adrenal axis

AB  - Dynamic properties of a nonlinear five-dimensional stoichiometric model of the hypothalamicpituitary-adrenal (HPA) axis were systematically investigated. Conditions under which qualitative transitions between dynamic states occur are determined by independently varying the rate constants of all reactions that constitute the model. Bifurcation types were further characterized using continuation algorithms and scale factor methods. Regions of bistability and transitions through supercritical Andronov-Hopf and saddle loop bifurcations were identified. Dynamic state analysis predicts that the HPA axis operates under basal (healthy) physiological conditions close to an Andronov-Hopf bifurcation. Dynamic properties of the stress-control axis have not been characterized experimentally, but modelling suggests that the proximity to a supercritical Andronov-Hopf bifurcation can give the HPA axis both, flexibility to respond to external stimuli and adjust to new conditions and stability, i.e., the capacity to return to the original dynamic state afterwards, which is essential for maintaining homeostasis. The analysis presented here reflects the properties of a low-dimensional model that succinctly describes neurochemical transformations underlying the HPA axis. However, the model accounts correctly for a number of experimentally observed properties of the stress-response axis. We therefore regard that the presented analysis is meaningful, showing how in silico investigations can be used to guide the experimentalists in understanding how the HPA axis activity changes under chronic disease and/or specific pharmacological manipulations.
