DNA protective activity of triterpenoids isolated from medicinal mushroom Fomitopsis betulina

AB  - Eleven 31-methylenlanostane triterpenoids, i.e., seven 21- and four 26-oic acids, as well as a lupane triterpenoid betulin, isolated from the fruiting bodies of the mushroom Fomitopsis betulina, were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. Most of the tested compounds showed a beneficial effect by reducing DNA damage of human lymphocytes more effectively than amifostine, a radioprotective agent, used as a positive control. All the tested compounds decreased MN frequency in the concentration dependent manner, with the concentration of 2.0 µg mL-1 being the most effective – with increase of the concentration the activity slightly decreases. The structure–activity relationship (SAR) studies indicated that the lanostanes containing a conjugated 7,9 (11)-diene system exhibit lower activity than Δ8-analogues. It was also demonstrated that the DNA protective activities within the Δ8-lanostane-26-oic acid group are affected by the substitution in position 3 pattern. In the Δ8 series the oxygenation at C-12 or 16 as well as 21- or 26-oic acid functionality proved beneficial for in vitro protective effect on chromosomal aberrations. Betulin exhibited the lowest protective activity, but it is still comparable to that of amifostine.
AB  - Једанаест 31-метиленланостанских тритерпеноида, то јест седам 21- и четири 26-
ланостанских киселина, као и лупански тритерпеноид бетулин изолованих из плодонос-
них тела гљиве Fomitopsis betulina тестирани су in vitro на заштитни ефекат на аберације
хромозома у периферним хуманим лимфоцитима. Примењен је тест мерења учес-
талости микронуклеуса индукованих применом инхибитора цитокинезе, цитохалазина
Б. Испитивања су показала да већина тестираних једињења показује значајан протек-
тивни ефекат на ДНК хуманих лимфоцита, већи него комерцијални радиопротективни
агенс аминофостин. У опсегу концентрација 1, 2 и 4 μg mL-1 сва испитивана једињења су
смањивала учесталост микронуклеуса (MN), при чему је најефиканија била концентра-

AB  - In continuation of our study on Cotinus coggygria from Serbia, 10 known flavonoids (1-10) were isolated from the methylene chloride/ methanol extract of the heartwood. They were tested for in vitro protective effect against chromosome aberrations in peripheral human lymphocytes, using the cytokinesis-block micronucleus assay. All tested compounds (in minimal doses of 1 μg/mL) exerted a beneficial effect by decreasing DNA damage of human lymphocytes in the range of 24.2% to 54.5%, better than the radio protectant control, amifostine. Functional groups, such as 3′,4′-dihydroxyphenyl (catechol), 5-OH, 3-OH, and 4-keto in flavonoids (3-keto in aurones), which play a key role in antioxidant activity, are proposed to be responsible for the DNA protective activity of the tested compounds.

AB  - Two sets of structurally different jatrophanes (1-11 and 13-16), jatrophane 12, and latex extract of 2 Euphorbia species (17 and 18) were tested for in vitro protective effect against chromosome aberrations in peripheral human lymphocytes using the cytokinesis-block micronucleus (CBMN) assay. Jatrophanes 1-6 in minimal doses of 1 µg/mL prominently decreased micronuclei (MN) frequency in the range 44.86% to 34.29% and manifested considerable protective effect. From the other set of jatrophanes, 13 in the same minimal dose notably decreased MN frequency by 31.05%, while extracts 17 and 18 at a concentration of 4 µg/mL remarkably decreased the frequency of MN by 37.94% and 36.12%, respectively. Jatrophanes 12, 14, and 16 showed moderate protection, while 7-11 and 15 were less active than positive control. The structure-activity relationship (SAR) studies of the tested jatrophanes (1-16) indicated the favorable position of benzoate at C-8 or C-9 (3, 4, and 13) and a preference of isobutanoyloxy group at C-3 (1-3) rather than propanoyloxy at the same position (4-6) for pronounced protective effect on human lymphocytes DNA. In a previous SAR study on 11 jatrophanes (1, 3-8, and 13-16), the same structural features in 3, 4, and 13 influenced powerful inhibition of P-gp, while growth inhibition of cancer cells was more than doubled in 1 (isobutanoyloxy group at C-3) compared to 6 (propanoyloxy at C-3).

Phenolic Compounds from Atriplex littoralis and Their Radiation-Mitigating Activity

AB  - From the aerial parts of Atriplex littoralis, three new flavonoid glycosides named atriplexins I-III (1-3), a known flavonoid glycoside, spinacetin 3-O-beta-D-glucopyranoside (4), arbutin (5), and 4-hydroxybenzyl-beta-D-glucopyranoside (6) were isolated. Their structures were elucidated on the basis of detailed spectroscopic analysis, including ID and 2D NMR (COSY, NOESY, TOCSY, HSQC, HMBC) and HRESITOF MS data. The compounds were tested for in vitro protective effects on chromosome aberrations in peripheral human lymphocytes using a cytochalasin-B-blocked micronucleus (MN) assay in a concentration range of 0.8-7.4 mu M of final culture solution. Chromosomal damage was induced by 2 Gy of gamma-radiation on binudeated human lymphocytes, and the effects of the compounds were tested 2 to 19 h after irradiation. The frequency of micronuclei (MNi) was scored in binudeated cells, and the nuclear proliferation index was calculated. The highest prevention of in vitro biochemical and cytogenetic damage of human lymphocytes induced by gamma-radiation was exhibited by 3 (reduction of MN frequency by 31%), followed by 4 and 6.

Their structures were elucidated on the basis of detailed spectroscopic analysis, including ID and 2D NMR (COSY, NOESY, TOCSY, HSQC, HMBC) and HRESITOF MS data. The compounds were tested for in vitro protective effects on chromosome aberrations in peripheral human lymphocytes using a cytochalasin-B-blocked micronucleus (MN) assay in a concentration range of 0.8-7.4 mu M of final culture solution. Chromosomal damage was induced by 2 Gy of gamma-radiation on binudeated human lymphocytes, and the effects of the compounds were tested 2 to 19 h after irradiation. The frequency of micronuclei (MNi) was scored in binudeated cells, and the nuclear proliferation index was calculated. The highest prevention of in vitro biochemical and cytogenetic damage of human lymphocytes induced by gamma-radiation was exhibited by 3 (reduction of MN frequency by 31%), followed by 4 and 6.",

AB  - Nine diarylheptanoids, 1-9, catechin (11), and a phenolic glucoside, 10, were isolated from the bark of green alder (Alnus viridis). Four of the isolated compounds, i.e., 2, 5, 8, 10, are new. The structures of 1-11 were determined on the basis of spectroscopic data. All isolated compounds were evaluated for their in vitro protective effects on chromosome aberrations in peripheral human lymphocytes using cytokinesis-block micronucleus (CBMN) assay. Almost all of them exerted a pronounced effect of decreasing DNA damage of human lymphocytes, acting stronger than the known synthetic protector amifostine.

AB  - This article summarizes the results of using cytokinesis-block micronucleus (CBMN) assay to evaluate the antioxidant potential of heterocyclic compounds. Most studies were carried out with naturally occurring heterocyclic compounds such as plant polyphenols: flavonoids, xanthones, coumarins, and ellagitannins, or plant derived products (juices, extracts, supplements) rich in bioactive heterocyclic compounds. There are also some studies dealing with synthetic heterocyclic antioxidants. CBMN assay is an in vitro study that has been used to evaluate antioxidant and protective effects of heterocyclic compounds on induced chromosome aberration in human lymphocytes.

AB  - On the basis of LC/UV/MS analysis, 35 compounds from the extracts of seeds of black, red, and white currants (2 cultivars of each) were identified. Black currants cultivars contained protocatechuic acid and p-hydroxybenzoic acid, and traces of nitril containing phenolic acids. The presence of synapoyl glucoside was characteristic for cv. Malling Jewel. Sesquiterpenoid glycosides and carboxymethylindol glycosides were present mainly in white and red currant cultivars. Blackcurrant seeds contained higher amounts of flavonoids, especially rutin, isoquercetin, and taxifolin. The currant seed extracts were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. The frequency of MN was scored in binucleated cells, and the cultivars of black currants showed the best antioxidant potential. At a concentration of only 0.17 mg/mL, extract of the seed cv. Malling Jewel effected a decrease in the frequency of MN of 60% compared with control cell cultures. Practical Application: Our results provide evidence of protective effects of currant seed extracts and isolated pure compounds on cytogenetic damages in human lymphocytes. Thus, currant seed extracts could exert beneficial effects in quite a few diseases, for many of the biological actions have been attributed to their antioxidant properties.

The BSEs and 3 isolated pure compounds were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (MN) assay. The frequency of MN was scored in binucleated cells, and nuclear proliferation index was calculated. Among the tested extracts, the seeds of cv. Thornfree at concentration of 1 mu g/mL exhibit the most prominent effect decreasing the frequency of MN by 62.4%, when compared with the controls cell cultures. Antioxidant potential of pure ellagitannins cannot explain the strong effect of BSEs. The assumption was that better antioxidant effect of BSEs result from synergistic effects of individual compounds contained in the extracts and/or some minor components possessed strong activity.

AB  - The chemical and sensorial characteristics of cherry brandy produced from five cherry varieties (Oblacinska, Celery's 16, Rexle, Heiman's Ruby and Heiman's Conserve) grown in Serbia were studied. Gas chromatography and gas chromatography-mass spectrometry analysis of these distillates led to the identification of 32 components, including 20 esters, benzaldehyde, 6 terpenes and 5 acids. The ethyl esters of C8-C18 acids were the most abundant in all samples. The benzaldehyde content was quantified by high performance liquid chromatography with UV detection. The average benzaldehyde concentration in the samples ranged between 2.1 and 24.1 mg L-1. The total sensory scores of the cherry brandies ranged between 17.30 to 18.05, with the cherry brandy produced from the Celery's 16 variety receiving the highest score (18.05).

AB  - This study was conducted to elucidate the in vitro protective effect of five flavonoids [apigenin (1), apigenin-7-O-glucoside (2), centaureidin (3), jaceidin (4) and quercetin (5)] against chromosomal damage in mitogen-induced human lymphocytes. Using the Cytochalasin-B blocked micronucleus (CBMN) assay, in which the biomarker of chromosome breakage and/or chromosome loss is the elevated frequency of micronucleus (MN) in binucleated (BN) cells, the presence of flavonoid 2 in minimal concentration (3 mu g/mL) gave a 35.5% decrease M the frequency of MN when compared with control human lymphocytes. The same concentration of flavonoids 1, 3 and 4, reduced the MN frequency by 24.4%, 28.0% and 28.0%, respectively. Higher concentrations (6 mu g/mL and 10 mu g/mL) seemed less effective. Flavonoid 5 (3 mu g/mL.) induced a slight decrease in MN frequency (5%), while higher doses (6 mu g/mL and 10 mu g/mL) provoked an increase of DNA damage. The comparable values for the cytokinesis-block proliferation index (CBPI) of the tested flavonoids and positive control suggested an inhibitory effect on lymphocyte proliferation. In the DPPH scavenging assay, flavonoids 1-4 demonstrated modest activity, in a dose-dependent manner, compared with the synthetic antioxidants BHT and Trolox, while 5 exhibited comparably high antioxidative activity.

AB  - A new prenylated flavanonol named seselinonol (1) was isolated from the roots of Seseli annuum, together with the well-known biologically active polyacetylenes falcarinol (2) and falcarindiol (3), and the prenylated furanocoumarin phellopterin (4). Its structure was elucidated by extensive spectroscopic analysis, including HR-ESI-MS, ID- and 2D-NMR. Seselinonol and phellopterin were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. The new compound exerted a beneficial effect by decreasing DNA damage of human lymphocytes.

AB  - In this study, volatile compounds were analyzed in five samples of home-made spirit beverage made by the distillation of fermented fruits of cornelian cherry (Cornus mas L.). The major volatile compounds, besides ethanol, identified and quantified were: methanol, acetaldehyde, I-propanol, ethyl acetate, 2-methyl-1-propanol, 1-butanol, amyl alcohols, 1-hexanol and 2-phenyl-ethanol, The minor volatiles were submitted to liquid liquid extraction with dichloromethane and analyzed by gas chromatography and gas chromatography/ /mass spectrometry (GC/MS). A total of 84 compounds were identified. The most abundant compounds were straight-chain free fatty acids, ethyl esters of C(6)-C(18) acids, limonene, 2-phenylethanol and 4-ethylphenol. Most of the compounds found in the "Drenja" spirits investigated in this study arc similar to those present in other alcoholic beverages.
AB  - U radu su analizirane isparljive komponente u pet uzoraka alkoholnih pića, domaće izrade, dobijenih destilacijom fermentisanih plodova drenjine (Cornus mas L.). Glavna isparljiva jedinjenja, koja su određena i kvantifikovana, su: metanol, 1-propanol, etil-acetat, 2-metil-1-propanol, 1-butanol, izoamil-alkoholi, 1-heksanol i 2-feniletanol. Manje zastupljena isparljiva jedinjenja su ekstrahovana dihlormetanom i analizirana kombinacijom gasne hromatografije i masene spektrometrije (GC/MS). Identifikovana su ukupno 84 jedinjenja. Najzastupljenija jedinjenja bila su etil estri masnih kiselina sa C6-C18, slobodne masne kiseline, limonen, 2-feniletanol i 4-etilfenol. Većina jedinjenja nađena u 'drenji' su slična onima koja su prisutna i u drugim alkoholnim pićima.

Antioxidant properties of raspberry seed extracts on micronucleus distribution in peripheral blood lymphocytes

AB  - This study addresses in vitro effects of raspberry (Rubus idaeus) seed extracts (RSE) on the frequency of micronuclei. We evaluated the effects of three different extracts (50%, 80%, and 100% methanol) in doses of 1.4, 4.2, and 8.4 mu g/mL, per 5 mL culture using cytochalasin-B micronucleus (CBMN) assay in peripheral human lymphocytes. The frequency of MN was scored in binucleated (BN) cells. The nuclear proliferation index was also calculated. The distribution of polyphenolic compounds in RSEs was determined using LC/UV/ESI-TOF MS. The identified 37 compounds comprised flavanol monomers and oligomers, as well as varieties of ellagitannin components. Treatment of lymphocytes with RSEs induced a significant decrease in the frequency of micronuclei by 80%. These results demonstrate that the constituents of RSEs may be important in the prevention of oxidative lymphocyte damage by reactive oxygen species and may also reduce the level of DNA damage. These findings support the potential benefits of polyphenolic compounds from raspberry seeds as efficient antioxidants.

Treatment of lymphocytes with RSEs induced a significant decrease in the frequency of micronuclei by 80%. These results demonstrate that the constituents of RSEs may be important in the prevention of oxidative lymphocyte damage by reactive oxygen species and may also reduce the level of DNA damage. These findings support the potential benefits of polyphenolic compounds from raspberry seeds as efficient antioxidants.",

AB  - The distribution of polyphenolic compounds in a grape (Vitis vinifera) seed extract (GSE) was determined using LC/ESI-TOF MS, HPLC/DAD, and C-13-NMR. The 17 identified compounds comprised gallic and protocatechuic acid, catetchin and epicatechin monomers, procyanidin oligomers, and procyanidin gallates. This study addresses the in vitro effects of grape seed extract (GSE) on the frequency of micronuclei with reference to the antioxidant status in human lymphocytes. To establish the most effective protective support, we used four different concentrations of GSE, in the range 1-6 mu g/mL. Treatment of lymphocytes with GSE at a concentration of 2.5 mu g/mL induced a significant decrease in the frequency of micronuclei by 40%, reduction of malonyldialdehyde production by 30%, while a concentration of 5 mu g/mL increased catalase and glutathione S-transferase activity by 10 % and 15 %, respectively. These results demonstrate that GSE may be effective in the prevention of oxidative lymphocyte damage by ROS.
