Antipsychotic Drug-Mediated Adverse Effects on Rat Testicles May Be Caused by Altered Redox and Hormonal Homeostasis

AB  - Sexual dysfunction, as a noticeable adverse effect of atypical antipsychotic drugs (APDs)
for the treatment of schizophrenia, has not been investigated in detail. A study was undertaken
to investigate whether 28-day long treatment with clozapine, ziprasidone or sertindole (using a
recommended daily dose for atypical antipsychotic therapy), induced histopathological changes
both in rat testicles and prostate, changed the activity of the antioxidant defence system and altered blood testosterone and prolactin. Clozapine, ziprasidone and sertindole induced histopathological changes in rat testicular tissue, which could be attributed to a disturbed testicular antioxidant defence system in addition to an altered prolactin to testosterone ratio. None of the APD treatments induced histopathological changes in prostate. Our results demonstrate that APDs have the capacity to change both redox and endocrinological balance. One or both outcomes could underline testicular degeneration and disturbed spermatogenesis.

AB  - Chronic use of atypical antipsychotics may produce hepatic damage. Atypical antipsychotics, including clozapine, sertindole, and ziprasidone, are extensively metabolized by the liver and this process generates toxic-free radical metabolic intermediates which may contribute to liver damage. The aim of this study was to investigate whether clozapine, sertindole, or ziprasidone affected hepatic antioxidant defense enzymes which consequently led to disturbed redox homeostasis. The expression and activity of antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR), catalase (CAT), and glutathione-S-transferases (GST) were measured in rat livers at doses corresponding to human antipsychotic therapy. Clozapine increased activity of SOD types 1 and 2, GR and GST, but reduced CAT activity. Sertindole elevated activities of both SODs. In ziprasidone-treated rats only decreased CAT activity was found. All three antipsychotics produced mild-to-moderate hepatic histopathological changes categorized as regenerative alterations. No apparent signs of immune cell infiltration, microvesicular or macrovesicular fatty change, or hepatocytes in mitosis were observed. In conclusion, a 4-week long daily treatment with clozapine, sertindole, or ziprasidone altered hepatic antioxidant enzyme activities and induced histopathological changes in liver. The most severe alterations were noted in clozapine-treated rats. Data indicate that redox disturbances may contribute to liver dysfunction after long-term atypical antipsychotic drug treatment.

AB  - Radicava™ (Edaravone) was approved the Food and Drug Administration (FDA) as a new treatment for amyotrophic lateral sclerosis (ALS). Edaravone is a synthetic antioxidant that specifically targets oxidative damage interacting with lipid radicals in the cell. In ALS disease the multiple cell types are involved in devastating loss of motor neurons. Mutations and biochemical changes in various cell types jointly contribute to motor neuron death, disease onset, and disease progression. The overall mechanism of neurodegeneration in ALS is still not completely understood. Dying motor neurons have been reported to exhibit features of apoptosis. However, non-apoptotic features of dying motor neurons have also been reported such as ferroptosis. The role of Edaravone in the prevention of ferroptosis in parallel with other therapeutic approaches to ALS therapy is discussed.

AB  - Objective: Early-onset and exclusive breast-feeding provides a significant health benefit to infants compared with infant formulas. The aim of this study was to compare mature breast milk with standard infant formulas by examining their effects on non-vascular smooth muscle contraction and their antioxidative properties. Methods: The pharmacologic effects of breast milk and formulas were examined using a model system of the rat uterine smooth muscle contraction. Electron paramagnetic resonance spin-trapping spectroscopy was used to compare the antioxidative capacities of breast milk (obtained in the ninth week of lactation) with commercial infant formulas against hydroxyl radical production in the Fenton reaction. The activities of superoxide dismutase, glutathione peroxidase, and the sulfhydryl group were determined in the breast milk and infant formulas. Results: In contrast to the infant formulas, breast milk exerted a relaxing effect on isolated non-vascular smooth muscle. In general, breast milk showed higher antioxidative activity compared with the infant formulas. In all samples, the generation of hydroxyl radicals led to the formation of carbon-centered and ascorbyl radicals. Conclusions: Human milk exerts direct pharmacologic relaxation effects and provides better antioxidant protection compared with infant formulas because of the presence of specific enzymatic components, such as human superoxide dismutase. We propose that these effects should be advantageous to an infant's gastrointestinal tract by supporting the normal work of the smooth musculature and maintaining redox homeostasis and may represent one of the mechanisms by which breast-feeding benefits health.

AB  - Early onset and exclusive breastfeeding provides a significant health benefit to infants in comparison to infant formulas. There is groving evidence that presence of some specific protein components in mothers milk are responsible for observed significant health benefit in infants feed with mothers milk. The aim of this paper was to compare mature breast milk and a standard infant formula by examining their effects on smooth muscle contraction and their antioxidative properties. Electron paramagnetic resonance (EPR) spin-trapping spectroscopy was used to compare the antioxidative capacities of breast milk (obtained on the 9th week of lactation) with a commercial infant formula against hydroxyl radical production in the Fenton reaction. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and their vitamin C and sulfhydryl group (–SH) contents were determined in the milks. Pharmacological research was performed on the isolated rat uterus. In contrast to the infant formula, breast milk exerted a
relaxing effect on isolated non-vascular smooth muscle. Using EPR and the Fenton reaction as a radical-generating system, we showed that breast milk possesses a three-fold higher antioxidative activity against the hydroxyl radical compared to the infant formula. In both samples, generation of the hydroxyl radical (•OH) led to the production of carbon-centered radicals. The ascorbyl radical was detected in breast milk but not in the infant formula. Human milk has direct pharmacological effects and provides better antioxidant protection than the infant formula due to the presence of specific protein components such as human SOD.

AB  - The conversion of nitric oxide (NO
) into its congeners nitrosonium (NO ) and nitroxyl (HNO/NO) ions may have important consequences for signal transduction and physiological responses. Manganese-containing superoxide dismutase (MnSOD) may convert
NO into its redox congeners. In our current work, we have examined the mechanism of sodium nitroprusside (SNP)-induced relaxation of arteries, with or without endothelium, from both normotensive and spontaneously hypertensive (SH) rats in the absence and
presence of MnSOD. SNP induced a greater degree of relaxation in normotensive than in SH rats. MnSOD antagonized SNPinduced relaxation and effect was greater in normotensive than hypertensive rats. However, MnSOD even potentiated SNP-induced
relaxation in mesenteric arteries with endothelium from SH rats. Our results indicate that HNO/NO-mediated relaxation is more effective in mesenteric artery smooth muscle from SH rats than from normotensive rats and that vascular dysfunction in SH rats is not

Effect of MnSOD (E. coli) on the relaxation caused by sodium nitroprusside on isolated rat renal artery

AB  - In this study themolecular foundation of nitric oxide induced relaxation of arteries, with or without endothelium, of normotensive and spontanously hypertensive ratswas re-examined. With this purpose in mind, the effects of the nitric oxide donor sodium nitroprusside (NaNP), with and without manganese containing superoxide dismutase (MnSOD E.C. 1.15.1.1), on rat renal artery relaxation was strudied. The results show that the relaxation effect of NaNP is two times higher in normotensive, compared to spontaneously hypertensive rats. Similar differences exist in the relaxation effects of NaNP on isolated renal arteries without endothelium, indicating that besides the difference in the function of an endothelium, concerning basal NO production in normotensive and hypertensive rats, there is a differencewith respect to NO relaxation in the smoothmuscle that is induced by hypertension. MnSOD decreased the relaxation effect of NaNP in all the examined renal arteries, more in normotensive than in hypertensive ones regardless of the presence of an endothelium. These results show that MnSOD, by modifying the chemical versatility of NO into redox active forms - nitrosonium (NO+) and nitroxyl (NO-), produces different relaxation effects in normotensive and hypertensive arteries of rats, with or without an endothelium, potentiating the role of nitroxyl induced relaxation in sponteneously hypertensive rats. The results prove the need for the synthesis of complex NO donors, as the mechanisms of artery relaxation are different due to an endothel and smooth mouscle changes in hypertensive, as compared to normotensive rats.
AB  - U ovom radu smo pokušali da detaljnije ispitamo molekulsku osnovu azotoksid indukovane relaksacije arterija, sa i bez endotela, normotenzivnih i spontano hipertenzivnih pacova. U tu svrhu ispitivan je efekat azot-oksid (NO) donora natrijum- nitroprusida (NaNP), bez i u prisustvu superoksid-dismutaze koja sadrži mangan (MnSOD, EC 1.15.1.1) na relaksaciju renalne arterije. Rezulati pokazuju dvostruko veći relaksantni efekat NaNP kod normotenzivnih, u odnosu na spontano hipertenzivne pacove. Slična razlika postoji i u relaksantnom efektu NaNP na izolovane renalne arterije normotenzivnog i hipertenzivnog pacova kada je odstranjen endotel što ukazuje da, pored razlike u funciji endotela u odnosu na bazalnu NO produkciju kod normotenzivnih i spontano hipertenzivnih pacova, postoje i promene u glatkim mišićima indukovane hipertenzijom, u odnosu na NO relaksaciju. MnSOD kod svih grupa smanjuje relaksantni efekat SNP i to više kod normotenzivnih nego kod hipertenzivnih nezavisno od prisustva endotela. Ovi rezultati pokazuju da MnSOD menjajući hemijsku prirodu iz NaNP oslobođenog NO u redoks aktivne forme – nitrozonijum (NO+) i nitroksil (NO–) – ostvaruje različit relaksantni efekat kod normotenzivnih i kod hiperetenzivnih pacova sa i bez endotela potencirajući značaj nitroksilom indukovane relaksacije kod spontano-hipertenzivnih pacova.
