A method for spectrophotometric determination of acetaldehyde with thiosemicarbazide in blood

AB  - The  thiosemicarbazide  method  for  spectrophotometric  determination  of  acetaldehyde    (AcH)    in    whole    blood    is    presented.    The    concentration    of    thiosemicarbazone  formed  in  slightly  acidic  medium  was  determined  at  260  nm.  The  influence of non-specific chromogens present in blood was eliminated by simultaneous AcH  distillation  and  thiosemicarbazone  formation.  The  oxidation  of  AcH  in  blood  samples  was  prevented  by  chloral  hydrate  addition.  The  presented  method  was  found  two   folds   more   sensitive   than   a   spectrophotometric   determination   of   AcH   by   semicarbazide.  Due  to  its  simplicity  and  sensitivity  as  well  as  to  the  use  of  low  cost  equipment,  this  method  should  be  a  valuable  complement  to  expensive  GLC  or  HPLC  determinations of AcH.
AB  - U radu je opisana metoda za spektrofotometrijsko određivanje acetaldehida (AcH) u krvi. Kao
reagens upotrebljen je tiosemikarbazid a apsorpcija nastalih u slabo kiseloj sredini
tiosemikarbazona određuje se na 260 nm. Uticaj nespecifičnih hromogena prisutnih u krvi
eliminiše se destilacijom AcH sa istovremenim građenjem tiosemikarbazona. Oksidacija AcH u
uzorcima krvi sprečena je dodatkom hloral hidrata. Opisana metoda se pokazala osetljivom u
odnosu na spektrofotometrijsko određivanje AcH sa semikarbazidom. Zbog svoje jednostavnosti,
osetljivosti i ekonomičnosti ova metoda može predstavljati korisnu zamenu za GLC i HPLC
određivanja AcH.

AB  - Insulinomas are pancreatic tumors associated with hyperinsulinism and hypoglycaemia. The effects of long-term hypoglycaemia, caused by hyperinsulinism in insulinoma patients, on erythrocyte metabolism, were in­vestigated. The modifications of erythrocyte metabolism were studied in prolonged hypoglycaemia. “Fast” haemoglobin modifications were re-invcstigated and the activi­ties in some erythrocyte enzymes of carbohydrate metab­olism and oxidative defense system, were determined. On the basis of the results obtained the structural alterations of haemoglobin molecule and the physiological impor­ tance of these changes, were assumed.

Anti-oxidative enzymes in pulmonary emphysemia

AB  - ln study anti-oxidative enzymes (Cat, G-Px, G-red, G-6PD11 and GSH) in patients with pulmonary emphysema, we analyzed the blood samples taken from 20 patients with global respiratory failure (10 males, mean age 56+-7.5 yis, and 10 females, mean age 60±5yrs). All patients were heavy cigarette smokers, in average 33±8 pack-years. Chest radiographs of these patients were compatible with lung emphysema. Spirometric examinations revealed severe, irreversible pressure-dependent airflow limitations; body plethysmographic measurements revealed overinflation in increased total lung capacities, and transfer factors were greatly reduced. The control group consisted of 14 healthy smokers. Slightly elevated (alpha-1-antitrypsin in our pts., 3.55±0.76 g/L, indicated an acute phase reaction. The highly positive correla­tion was found between Cu and LDH (r=0.81, p<0.001) and between Cu and Mg (r=0.47, p<0.01). These results are correlated to the parameters of acid - base status, and they suggest that it is necessary to determine copper in the blood of patients with COLD in order to correctly evaluate hypoxia.

The mean values of ceruloplasmin were similar in the two groups (0.45±0.01 ; 0.37±0.05 g/L). Scrum copper levels were also similar in both groups (126±46 : 134±14pg/100mL). The other authors have stated that in inflam­mation the copper concentration was increased. However our findings could not prove it. Decreased concentration of zinc pointed to the destabilization of lysosomal mem­ branes and increased macrophage activity that liberates proteases.

AB  - In this work the effect o f toluene exposition on oxidative chemical species generation, metabolic alteration and protective effect of selenium were investigated.
The disturbances in erythrocyte metabolism due to oxidative processes were studied by the use o f experimental animals. The alterations of some erythrocyte enzyme activities and antioxidative molecules were found in exposed to toluene. Also, the more intensive primary antioxidative response to oxidative stress and the protective effect of selenium was detected.

AB  - Za istraživanja acctaldchidnih adukata hemoglobina upotrcbljcni su liemolizati lironičnih alkoholičara, koji nisu bili pod terapijom i hemolizati zdravih osoba radi poredenja. Pored toga analizirana su i acetaldchidna jedinjenja hemoglobina sintetizovana inkubiranjem hemolizata zdravih osoba kao ifrakcija HbAg i HbAj, sa acctaldehidom pod uslovima in vitro. Da bi se ustanovio mogući uticaj drugih biomolekula na vezivanje acetaldehida hemolizati su inkubirani sa acctaldehidom, sa glukozom, fosforilovanim šećerima i oksidovanim glutationom. Analiza jc u svim slučajevima vršena primenom jonoizmenjivačkih i clcktroforetskih tclinika a u odvojenim frakcijama određen jc sadržaj kovalentno vezanih ugljenih hidrata, acetaldehida i slobodnih -SH grupa spektrofotometrijski. Da bi se utvrdio sadržaj "nestabilnih" acctaldchidnih adukata u svim slučajevima izvedeni su i ogledi i prisustvu NaBH4 . Na osnovu dobivenih rezultata zaključilo se da su "brze" hcmoglobinskc frakcije (HbA0 povećane kod alkoholičara (20%). U ovoj frakciji dokazan jc veći broj hemoglobinskih modifikacija u odnosu na zdrave osobe. U većini izdvojenih frakcija dokazan jc za hemoglobin vezani acctaldcliid kao i prisustvo "nestabilnih" adukata.
Zaključeno jc da sc in vivo u hemolizatima alkoholičara grade više vrsta acctaldeliidnih adukata sa hemoglobinom različite stabilnosti. Potvrde o pretpostavljenim mestima i načinu vezivanja acetaldehida za hemoglobin dobivene su u ogledima in vitro inkubiranjem hemolizata, HbA0 i HbAj frakcije sa različitim biomolckulima i acctaldehidom. U ovim ogledima utvrđen jc i inliibitomi uticaj nekih biomolekula na vezivanje acctaldcliida. Broj i mesta vezivanja po lancima utvrđen jc analizom odvojenih hemoglobinskih lanaca, pri čemu jc dokazano više tipova licmoglobinskih modifikacija. Uticaj vezivanja acetaldehida na kvatemernu strukturu hemoglobina dokazan je utvrđenim većim stepenom disocijacijc u frakcijama sa većim sadržajem acetaldehida. Iz svega se zaključilo, da se kako pod uslovima in vivo tako i in vitro pod uticajcm acetaldehida grade više tipova hemoglobinskih acctaldchidnih adukata, i koji utiču na kvaternemu strukturu hemoglobina, i njegovu funkciju i mogu biti od patofiziološkog značaja.

Spektrofotometrijsko određivanje acetaldehida u biološkom materijalu

AB  - Za analizu korišćeni su uzorci venske krvi pripremljenih sa dodatkom ACĐ rastvora i hloralhidrata i do upotrebe čuvani na <4°C. Standardni rastvori acetaldehida su pripremljeni razblaživanjem standardnog rastvora acetaldehida koncentracije 780g/L. Acetaldehid je izdvojen destilacijom u puferovani rastvor tiosemikarbazida.
Koncentracija nagrađenog tiosemikarbazona određena je merenjcrn absorbance na 258 nm. Lspitivanjanjcm određeni su najoptimalniji uslovi za primenu metode kod direktne reakcije tiosemikarbazida sa acctaldehidom i to: pH reakcione sredine (plI6), koncentracija tiosemikarbazida (0,004M), vreme odvijanja reakcije (20min.).
Ispitivani su uslovi reakcije tiosemikarbazida sa acetaldehidom, izdvojenog destilacijom. Ova ispitivanja su posebno značajna jer se acetaldehid nalazi u krvi koja sadrži mnoge supstance koje bi mogle ometati određivanje pa je potrebno izdvajanje acetaldehida iz uzorka destilacijom ili nekom drugom pogodnom metodom. Pored toga ispitana je i postojanost tiosemikarbazidnog reagensa (30 dana na +4°C), postojanost acetaldehida u uzorku krvi (7dana +4°C uz dodatak hloralhidrata). Na kraju izvršeno je i upoređivanje osetljivosti i preciznosti tiosemikarbazidne metode u intervalu koncentracije acetaldehida od 0,1-1,0 mg/L. Metoda je proverena primenom za određivanje kako slobodnog tako i za proteine vezanog acetaldehida. Iz dobivenih rezultata utvrđeno je da je predložena metoda osctljivija i preciznija pa se može upotrebiti za određivanje malih količina acetaldehida u biološkom materijalu složenog sastava. Metoda se pokazala dovoljno specifičnom da zameni skupa i složena, enzimska, GLC i HPLC određivanja.

AB  - Stepen glikemije može biti određen analizom sadržaja glukoze ili ugljenih hidrata ne-enzimski veza­ nih za proteine krvi. Već se duže vremena koristi određivanje ugljenih hidrata vezanih za hemoglobulin ili proteine seruma. Za ova određivanja koriste se, pored ostalih, najčešće spektrofotometrijske metode sa tiobarbiturnom kiselinom kao reagensom za izazi­ vanje boja. Međutim ova metoda se pokazala relativno neosetljivom i nereproduktivnom. Cl ovome radu data je nova spektrofotometrijska metoda za određivanje ne-enzimski vezanih ugljenih hidrata za proteine seru­ ma, koja se zasniva na reakciji sa fenol-sumpornom kiselinom. Ispitani su i značajni parametri za primenu metode, kao što su uslovi izvođenja hidrolize, građenja hidroksimetil-furfurala i hromofore, kao i mogući ne­ specifični uticaj na reakciju. Metoda se pokazala više osetljivom od spektrofotometrijskog određivanja sa tiobarbituralnom kiselinom. Nađena je bolja korelacija sa sadržajem HbAic određenim jonoizmenjivačkom hromatografijom, povećanje sadržaja ugljenih hidrata kod dijabetičara bilo je značajnije. Pored toga nije ustanovljen nespecifičan uticaj drugoih supstanci na određivanje.
AB  - The degree of glycemic control may be performed by analysis of serum glycose content, as well as by determination of non-enzymatically protein-bound carbohydrates in blood. Thus, the determination of hemoglobin and serum protein-bound carbohydrates are used in clinical praxis. These determinations were performed commonly by spectrophotometric method based on thiobarbituric acid as reagent. But this method is not sensitive and reproductive enough. In this work a new spectrophotometric method for determination of non-enzymatically serum-protein bound carbohydrates was proposed. In the method phenol-sulfuric acid as reagent was used. All relevant parameters were investigated, as the conditions of hydrolysis and formation hydroxymethyl-furfural, as well as non-specific influences on the reaction. The method was found more sensitive than spectrophoto-metric one with use thiobarbituric acid as reagent. More significant correlation with HbAtc contents, determined by ion-exchange chromatography, was found. The elevation of protein-bound carbohydrates in diabetics was more significant and the non-specific influences on the determination was not found.

Uticaj aromatičnih organskih rastvarača na antioksidativni sistem u eritrocitima

AB  - Slobodni kiseonični radikali su izuzetno reaktivan hemijska jedinjenja. Kao takvi lako mogu dovesti do oštećenja različitih biomolekula enzima, ćelijskih mem­ brana, pa im se pripisuje potencijalna uloga u genezi mnogih patoloških procesa. Izlaganje dejstvu aroma­ tičnih organskih rastvarača može dovesti do stvaranja većih količina slobodnih kiseoničnih radikala. U ovome radu ispitivan je efekat izlaganja dejstvu aromatičnih organskih rastvarača i slobodnih kiseoničnih radikala na metabolizam u eritrocitima. Analizirani su indeksi antioksidativnog statusa eritrocita, sadržaj glutationa i antioksidativnih enzima (glutationperoksidaze i reduk- taze, katalaze i superoksidne dizmutaze). Takođe je ispitan efekat oskidativnih radikala na metabolizam hemoglobina i ugljenih hidrata. Određena je aktivnost methemoglobin reduktaze i sadržaj hemoglobinskih derivata.
AB  - Free radicals are extremly reactive chemical species. Damaging various biomolecules, enzymes, cell membranes they have a potential role in many pathological processes. Exposition to aromatic organic solvents may lead to increase generation of free radicals. The effect of aromatic organic solvents and generated free radicals on erythrocyte metabolism are investigated. The indices of antioxidant status in erythrocytes, glutathione and antioxydant enzymes (glutathione peroxidase and reductase, catalase and superoxidismutase) were investigated. The effect of oxidative radicals upon hemoglobin and carbohydrate metabolism was investigated too. Hemoglobin modifications and derivatives, as well as activity of methemoglobin reductase was analysed as well as the activities of key anzyrnes of pentose phosphate shunt.

Kao takvi lako mogu dovesti do oštećenja različitih biomolekula enzima, ćelijskih mem­ brana, pa im se pripisuje potencijalna uloga u genezi mnogih patoloških procesa. Izlaganje dejstvu aroma­ tičnih organskih rastvarača može dovesti do stvaranja većih količina slobodnih kiseoničnih radikala. U ovome radu ispitivan je efekat izlaganja dejstvu aromatičnih organskih rastvarača i slobodnih kiseoničnih radikala na metabolizam u eritrocitima. Analizirani su indeksi antioksidativnog statusa eritrocita, sadržaj glutationa i antioksidativnih enzima (glutationperoksidaze i reduk- taze, katalaze i superoksidne dizmutaze). Takođe je ispitan efekat oskidativnih radikala na metabolizam hemoglobina i ugljenih hidrata. Određena je aktivnost methemoglobin reduktaze i sadržaj hemoglobinskih derivata., Free radicals are extremly reactive chemical species. Damaging various biomolecules, enzymes, cell membranes they have a potential role in many pathological processes. Exposition to aromatic organic solvents may lead to increase generation of free radicals. The effect of aromatic organic solvents and generated free radicals on erythrocyte metabolism are investigated. The indices of antioxidant status in erythrocytes, glutathione and antioxydant enzymes (glutathione peroxidase and reductase, catalase and superoxidismutase) were investigated. The effect of oxidative radicals upon hemoglobin and carbohydrate metabolism was investigated too. Hemoglobin modifications and derivatives, as well as activity of methemoglobin reductase was analysed as well as the activities of key anzyrnes of pentose phosphate shunt.",

AB  - Insulinoma, pancreas tumors lead to the increased insulin secretion, glucose consump­tion and the hypoglycemia emergence. The changes of erythrocyte metabolism in hypoglycemia have not been studied so far.
To explain the influence of hypoglycemia on erythrocyte metabolism the activities of erythrocyte enzymes and the content of hemo­globin derivates were analyzed. More precise identification of some modified hemoglobins was carried out; and their contents we­re analysed too.The physiological importance and practical application of results remains to be solved.
