Comparative Analysis of Phenolic Compounds in Seven Hypericum Species and Their Antioxidant Properties

AB  - A comparative analysis of the phenolic compounds in the 70% EtOH extracts of Hypericum acutum, H. androsaemum, H. barbatum, H hirsutum, H maculatum, and H richeri has been carried out using high-performance liquid chromatography coupled with photodiode array UV detection and high resolution TOF mass spectrometry. Quercetin, astilbin, I3,II8-biapigenin, orientin, 2"-O-acetylorientin, three phenolcarboxylic acids, and eight flavonols 3-O-glycosides were identified in the extracts on the basis of their on-line UV spectra, accurate mass spectral data, and in comparison of retention times with those from the standards. Fingerprint analysis of the extracts revealed significant differences in the qualitative and quantitative chemical composition of the studied species. Antioxidant assays with various reaction mechanisms were used including ferric reducing antioxidant power (FRAP) assay, DPPH, ABTS, superoxide anion radical scavenging capacity and inhibition of liposome peroxidation induced by Fe2+. The most potent were extracts of H acutum and H maculatum indicating this Hypericum species interesting for further research aimed as a potentially new source of biologically active compounds.

AB  - The chemical composition and free radical scavenging properties of the MeOH extract of Hypericum richeri and its subsequent CHCl3, n-BuOH, EtOAc and aqueous extracts were investigated. Four quercetin-3-O-glycosides, three myricetin-3-O-glycosides, 3-O- and 5-O-caffeoylquinic acid, quercetin, I3,II8-biapigenin, pseudohypericin and hypericin were identified. The constituents of the extracts were quantified using HPLC-DAD (high-performance liquid chromatography-diode array detector). Isolation of the compounds from n-BuOH and EtOAc extracts was conducted by column chromatography. Antioxidant capacity of the extracts was evaluated by superoxide radical and ABTS root+ scavenging assays and inhibition of lipid peroxidation. The EtOAc extract of H. richeri was the most potent antioxidant in all experiments. Free radical scavenging compounds were identified by HPLC-DPPH root (high-performance liquid chromatography-2,2-diphenyl-1-picrylhydrazyl) post-column derivatisation method, where all phenolic compounds except I3,II8-biapigenin exhibited radical scavenging properties. This is the first report of the presence of myricetin-3-O-rutinoside, myricetin-3-O-galactoside, myricetin-3-O-glucoside, 5-O-caffeoylquinic acid and I3,II8-biapigenin in H. richeri.

Anti-inflammatory and Gastroprotective Properties of Hypericum richeri Oil Extracts

AB  - Oil extracts of flowering tops of Hypericum richeri Vill. prepared in three different ways were evaluated for chemical composition, and anti-inflammatory and gastroprotective activities. An HPLC method was developed for determination of two dominant flavonoids, quercetin and I3,II8-biapigenin. The carrageenan-induced rat paw edema test was used for screening the anti-inflammatory activity, while indomethacin-induced rat gastric mucosa damage test was used for evaluation of gastroprotective activity. The oil extract prepared by maceration with 96% ethanol, followed by extraction with sunflower oil by heating on a water bath, exhibited the highest anti-inflammatory (38.4 %) and gastroprotective activities (gastric damage score of 0.9). The same oil extract had the highest content of quercetin (49 mu g/mL) and I3,II8-biapigenin (60 mu g/mL). These results approve the usage of oil extracts of H. richeri as an anti-inflammatory and gastroprotective agent.

Influence of thermal treatment on polyphenolic compounds and antioxidant properties of Quercus robur kernels

AB  - Polyphenolic compounds and antioxidant properties of dried MeOH extracts of native (QN) and thermally treated (QT) kernels of English Oak, Quercus robur were investigated. The content of total and simple polyphenols and tannins was determined spectrophotometrically using FC-reagentr, while gallic acid content was estimatecl usipg HPLC2. After thermal treatment tannin content was decreased (from 20,4 to 19,70 ), with significant increase of simple polyphenols and gallic acid content (from 1,9 to 4,004, and from 0,5 to 5,lYo, respectively). Antioxidant activity was estimated using FRAP test (total antioxidant activity, TAA), scavenging of DPPH radical and TBA test (inhibition of lipid peroxidation, LP in liposomes)r. Sample-s QN and QT showed substantial antioxidant activify. TAA was 2.44 and. 3.76 pmol F.'*/mg, respectively. Comparing to quercetin and L-ascorbic acid these values were ca 2-3 times lower. Both samples concentration-dependently scavenged stable DPPH radical (IC5s:8.04 and 5.77 pglml), and inhibited LP (IC50:32.13 and I 6.39 1tg/m| respectively). Higher antioxidant activity of extract of thermally treated kernels (QT) could be explained as a consequence of thermal degradation of hydrolysable tannins and, therefore, increased gallic acid content.

SadrZaj ukupnih i netaninskih polifenola i tanina odredivan je spektiofotometrijski, kori5cenjem Folin-Ciocalteu reagensa', a sadraaj galne kiseline tecnonr hromatografijom (HPLC)2. Konstatovano je da nakon termicktg tretmana sadrZaj tanina opada (sa 20,4 na 19,7o ), a znalajno raste kolidina netaninskih polifenola 1sa f ,9 na 4,0oA) i galne kiseline (sa 0,5 na 5 ,loh). Chemical Composition of Tanacetum larvatum Essential Oil

AB  - Tanacetum larvatum (Oris.) Kanitz. is an endemic perennial herb distributed on a rocky terrain in Serbia (Kosovo), Montenegro and Albania. The chemical composition of T. larvatum essential oil, obtained from two locations in Montenegro (Mt. Komovi [Sample I] and Mt. Prokletije [Sample II]) was investigated. The oils obtained by hydrodistillation were analyzed by GC and GC/MS, on the two different capillary columns. The aerial parts of T. larvatum yielded 0.3% and 0.2% of a yellowish oil (Sample I and Sample II, respectively). About 40 compounds were identified, representing ∼89% and 96% of the total oil content in the Samples I and II, respectively. trans-Sabinyl acetate was found to be the dominant component (51.2% and 69.7%). Among the rest of compounds β-pinene (7.7% and 4.3%) and camphor (6.3% and 4.3%) were the most abundant in both samples.

About 40 compounds were identified, representing ∼89% and 96% of the total oil content in the Samples I and II, respectively. trans-Sabinyl acetate was found to be the dominant component (51.2% and 69.7%). Among the rest of compounds β-pinene (7.7% and 4.3%) and camphor (6.3% and 4.3%) were the most abundant in both samples.",
