Assessment of the Biological Effects of Pellia endiviifolia and its Constituents in Vitro

AB  - Liverworts are characterized by a high content of bioactive compounds reported to show antimicrobial, anticancer, and antioxidant properties. In this study, the biological effects of the methanol extract of the liverwort Pellia endiviifolia and its constituents, bis-bibenzyls perrottetin E, 10′-hydroxyperrottetin E, and 10,10′-dihydroxyperrottetin E, were investigated using human peripheral blood cells as a model system. The assessment of the investigated compounds comprised testing their genotoxicity, apoptotic potential, and redox modulating activities. The genotoxicity testing indicated that medium (25 µM) and high concentrations (100 µM) of the investigated compounds displayed genotoxic and antiproliferative effects in human lymphocytes as revealed by significant, concentration-dependent enhancement of the micronuclei incidence and decrease in the cytokinesis-block proliferation index compared to the control (P <.001). Analysis of leukocyte apoptosis showed a substantial potential of all investigated compounds to induce apoptosis, which was not concentration-dependent. The P endiviifolia extract and perrottetin E demonstrated considerable pro-apoptotic potential, even at the lowest concentration (1 µM) applied. Evaluation of the redox modulating effects, which comprised measuring erythrocyte catalase activity and the lymphocyte malondialdehyde level, showed that the investigated compounds did not induce oxidative stress in human peripheral blood cells (P >.05). The observed genotoxic, antiproliferative, and proapoptotic effects of the investigated compounds make them suitable for further comprehensive studies related to their possible applications as anticancer agents.

AB  - Acetylcholinesterase (AChE) inhibitors are important in the treatment of neurodegenerative diseases. Two inhibitors,
12-tungstosilicic acid (WSiA) and 12-tungstophosphoric acid (WPA), which have polyoxometalate
(POM) type structure, have been shown to inhibit AChE activity in nM concentration. Circular dichroism and
tryptophan fluorescence spectroscopy demonstrated that the AChE inhibition was not accompanied by significant
changes in the secondary structure of the enzyme. The molecular docking approach has revealed a new
allosteric binding site, termed β-allosteric site (β-AS), which is considered responsible for the inhibition of AChE
by POMs. To the best of our knowledge, this is the first study reporting a new allosteric site that is considered
responsible for AChE inhibition by voluminous and negatively charged molecules such as POMs. The selected
POMs were further subjected to genotoxicity testing using human peripheral blood cells as a model system. It
was shown that WSiA and WPA induced a mild cytostatic but not genotoxic effects in human lymphocytes, which
indicates their potential to be used as medicinal drugs. The identification of non-toxic compounds capable of
binding to an allosteric site that so far has not been considered responsible for enzyme inhibition could be
fundamental for the development of new drug design strategies and the discovery of more efficient AChE
modulators.

AB  - U ovom radu ispitivan je uticaj gama zračenja na molekulsku masu i staklasti prelaz poli-L-laktida (PLLA). S obzirom da je PLLA pri izlaganju visokoenergijskom zračenju sklon kidanju lanaca i velikoj degradaciji, promene u molekulskoj masi su određene primenom gel propusne hromatografije (GPC). Radijaciono-indukovane promene u staklastom prelazu su ispitane pomoću diferencijalno skenirajuće kalorimetrije (DSC). Prividna aktivaciona energija za staklasti prelaz, ΔH*, određena je na osnovu zavisnosti temperature staklastog prelaza, Tg, od brzine zagrevanja. Kidanje lanaca, uzrokovano zračenjem, glavni je razlog opadanja Tg i ΔH* sa apsorbovanom dozom. Izazvane promene u molekulskoj masi i temperaturi staklastog prelaza odigravaju se na predvidljiv i prilično precizan način. Stoga, gama zračenje može biti korišćeno nesamo za sterilizaciju nego i za dobijanje ovog biomaterijala poželjnih karakteristika.
AB  - The influence of gamma radiation on the structure and glass transition behaviour of poly-L-lactide (PLLA) has been studied. Since PLLA exposed to high-energy radiation in the presence of air is prone to chain scission reactions and large degradation, changes in molecular weight were obtained by the gel permeation chromatography (GPC). Alterations in the glass transition behaviour were investigated by differential scanning calorimetry (DSC). The apparent activation energy, ΔH*, for the glass transition was determined on the basis of the heating rate dependence of the glass transition temperature, Tg. Our findings support the fact that chain scission is the main reason for the decrease of Tg and ΔH* with the absorbed dose. At low doses, despite large changes in molecular weight, only small alterations are observed in the glass transition temperature and apparent activation energy. Further increase in the absorbed dose introduces not only significant changes in the molecular weight but also in the glass transition temperature and the activation energy. Such glass transition behavior is a manifestation of a well-known effect of molecular weight on Tg, postulated by the Fox–Flory equation. However, all the observed alterations are small and tolerable at absorbed doses required for sterilization (up to 25 kGy in most circumstances). Furthermore, this study reveals that the radiation-induced changes in the molecular weight and the glass transition temperature occur in a predictable and fairly accurate manner. Therefore, gamma radiation can be used not only for sterilization but also for tailoring desirable end-use properties of PLLA based implants.

AB  - The influence of orientation on polyolefins was studied in accordance with Peterlin’s molecular model of drawing. Isotactic polypropylene and low density, linear low density and high density polyethylenes were oriented via solid-state stretching at an elevated temperature. Orientation induced changes in the morphology were analyzed by optical and scanning electron microscopy. Differential scanning calorimetry and wide-angle X-ray diffraction were used to determine changes in the crystallinity where a two-stage evolution was revealed. The first stage is characterized by a significant increase in crystallinity while the following saturation occurs with a further draw ratio increase at the second one. The value of the critical draw ratio, which separates these two stages and corresponds to the transformation from the initial to the fully developed fibrillar structure, was influenced by the structural peculiarities of each polyolefin.
