AB  - This study was performed to determine the main antibacterial compounds of the essential oil (ЕО) of saltmarsh plant Artemisia santonicum (Asteraceae). The combination of HPTLC and direct bioautography was used for the activity guided isolation of isogeranic acid as the main antibacterial constituent with remarkable antimicrobial activity, although it was the minor component of the EO, present only in 0.2 %, as calculated from GC/FID. Its structure was determined by 1D- and 2D-NMR and GC–MS techniques. Antibacterial activity of isogeranic acid against all tested bacteria was significantly higher than EO and even than both controls streptomycin and ampicillin. In further investigation of antibiofilm and antiquorum sensing activity EO exhibited the best inhibition of the biofilm formation at 1/8 minimal inhibitory concentration (MIC) and isogeranic acid at 1/2 MIC. Both EO and isogeranic acid possessed pyocyanin inhibitory activity showing the reduction of pigment at 60.6 and 62.8 %, respectively, at 1/2 MIC concentrations.
AB  - Ово истраживање је спроведено у циљу одређивања главних антибактеријских компоненти етарског уља слатинске биљне врсте Artemisia santonicum (Asteraceae). Комбинација HPTLC методе и методе директне биоаутографије је коришћена за активношћу вођено изоловање изогеранилне киселине као главне компоненте са значајном антимикробном, антибиофилм и антикворум активношћу иако је била присутна у етарском уљу са само 0,2 %, израчунато из GC/FID. Њена структура је одређена 1D и 2D NMR и GC–MS техникама. Антибактеријска активност изогеранилне киселине на све тестиране бактерије је била значајно боља од етарског уља, и од обе контроле, стрептомицина и ампицилина. Етарско уље је показало најбољу инхибицију формирања биофилма у концентрацији 1/8 минималне инфибиторске концнетрације (MIC вредности), а изогеранилна киселина у концентрацији 1/2 MIC вредности. И етарско уље и изогеранилна киселина су имале пиоцијанин инхибиторну активност показавши смањење пигмента на 60,6 и 62,8 %, у концентрацији 1/2 MIC вредности, редом.

Antibacterial and Antibiofilm Activity of Flavonoid and Saponin Derivatives from Atriplex tatarica against Pseudomonas aeruginosa

AB  - A new flavonoid glucoside derivative, patuletin 3-O-(2-O-feruloyl)-β-D-glucuronopyranosyl-(1→2)-β-Dglucopyranoside, named atriplexin IV (1), and three new triterpenoid saponin derivatives, two sulfonylated, β-Dglucopyranosyl-3-O-(2-O-sulfo-β-D-galactopyranosyl)-(1→2)-α-L arabinopyranoside-30-alolean-12-en-28-oate (2), named atriplexogenin I, β-D-glucopyranosyl-3-O-(2-O-sulfo-β-D-galactopyranosyl)-(1→2)-α-L-arabinopyranoside)-30-hydroxyolean-12-en-
28-oate (3), named atriplexogenin II, and β-D-glucopyranosyl-3-O-(β-D-glucopyranosyl-(1→2)-β-D-galactopyranosyl-(1→2)-α-L-arabinopyranoside)-30-alolean-12-en-28-oate (4), named atriplexogenin III, were isolated by silica gel column and semipreparative HPLC chromatography from the n-butanol extract of the salt marsh plant Atriplex tatarica. In addition, two known secondary metabolites, patuletin3-O-β-D-apiofuranosyl-(1‴→2″)-β-D-glucopyranoside (5) and patuletin 3-O-5‴-Oferuloyl-β-D-apiofuranosyl-(1‴→2″)-β-D-glucopyranoside (6), were isolated for the first time from A. tatarica. The structures of the isolated compounds were elucidated by 1D and 2D NMR, HRESIMS, IR, and UV data. Antibacterial activity by the microdilution method and antibiofilm activity against P. aeruginosa were assessed. Compound 5 possesses significant
antibacterial activity, while the most potent antibiofilm agent is compound 2.

Phenolic Compounds from Atriplex littoralis and Their Radiation-Mitigating Activity

AB  - From the aerial parts of Atriplex littoralis, three new flavonoid glycosides named atriplexins I-III (1-3), a known flavonoid glycoside, spinacetin 3-O-beta-D-glucopyranoside (4), arbutin (5), and 4-hydroxybenzyl-beta-D-glucopyranoside (6) were isolated. Their structures were elucidated on the basis of detailed spectroscopic analysis, including ID and 2D NMR (COSY, NOESY, TOCSY, HSQC, HMBC) and HRESITOF MS data. The compounds were tested for in vitro protective effects on chromosome aberrations in peripheral human lymphocytes using a cytochalasin-B-blocked micronucleus (MN) assay in a concentration range of 0.8-7.4 mu M of final culture solution. Chromosomal damage was induced by 2 Gy of gamma-radiation on binudeated human lymphocytes, and the effects of the compounds were tested 2 to 19 h after irradiation. The frequency of micronuclei (MNi) was scored in binudeated cells, and the nuclear proliferation index was calculated. The highest prevention of in vitro biochemical and cytogenetic damage of human lymphocytes induced by gamma-radiation was exhibited by 3 (reduction of MN frequency by 31%), followed by 4 and 6.

Screening inland halophytes from the central Balkan for their antioxidant activity in relation to total phenolic compounds and flavonoids: Are there any prospective medicinal plants?

AB  - This paper presents new data for secondary metabolites and biological activity of the insufficiently studied European inland halophytes as remarkable indicators of Balkan saline habitats. Phenolic content, flavonoid concentrations and antioxidant activity were analyzed in fifteen terrestrial salt tolerant plants collected from eight saline habitats situated in two distinct semiarid regions of Serbia. Total phenols ranged from 31.86 to 212.71 mg of GA/g of extract (gallic acid equivalent milligrams of gallic acid per gram of extract). Concentration of flavonoids ranged from 41.21 to 146.06 mg of RU/g of extract (rutin equivalent milligrams of rutin per gram of extract). Antioxidant activity was determined using 1,1-dipheny1-2-picrylhydrazyl radical reagent; obtained values was ranged from 1785.81 to 17.55 mu g/ml. A significant relation was noticed between the concentration of phenols and antioxidant activity. Parallel to the analysis of researched halophytes, Camellia sinensis and Ginkgo biloba were analyzed for comparison. The best antioxidant properties were determined for Statice gmelinii, Artemisia santonicum and Mentha pulegium. The results indicate a potential of studied halophytic species as candidates for natural sources of secondary metabolites with high biological activity, whereas their bioactive compounds might have a significant role in adaptations to salt and drought stress.

AB  - Arnica (Arnica montana L) is a perennial mountain plant whose flower heads are mainly used for topical treatment of bruises and sprains in phytopharmaceutical preparations such as ointments, creams and gels. In this study, different models of arnica cultivation have been tested in three-year field trial arranged in 2 x 3 x 2 factorial design. Planting date (spring and autumn), fertilizater type (manure, NPK and control) and mode of reproduction (generative and clonal) were induced factors on the trial. The variation of the rosette diameter, flowering stem count and height, flower heads count and diameter, together with flower heads yield and chemical properties were investigated as morphological parameters. Spring planting of clonally propagated plants was showed to be the least successful model due to the low percent of rooted plants and weak shoot development. Other models yielded from 59.8 to 143.6 kg ha(-1) and 116.3 to 258.7 kg ha(-1) flower heads (dw) in the second and third year, respectively. Total sesquiterpene lactones content in flower heads of all tested variants was above the minimum of pharmacopeia quality regulation and ranged from 4.6 to 13.9 mg g(-1) depending on factor levels, while the amounts of two dominant flavonoid compounds quercetin-3-O-glucoside and kaempferol-3-O-glucoside ranged from 7.8 to 13.9 mg g(-1) and from 2.1 to 4.7 mg g(-1), respectively. All induced factors in the field trial significantly influenced variation in both morphological and chemical parameters, where fertilized variants from autumn planting have shown to be superior in flower heads yield with satisfying levels of the secondary metabolites content. Factorial analysis distinguished that half of morphological parameters positively correlated with flower heads yield. First three ranks of sequential classification of cultivation models with desirable properties were captured by variants planted in autumn, while the last rated variants were those from clonal propagation planted in spring. This attempt of arnica domestication in agroecological conditions of western Serbia has proven to be successful in ensuring high quality raw material, providing at the same time answers to the questions that have not been reviewed so far.
