High-quality draft genome sequence of Pseudomonas aeruginosa san ai, an environmental isolate resistant to heavy metals

AB  - The strain Pseudomonas aeruginosa san ai, isolated from an extreme environment (industrial mineral cutting oil, pH 10), is able to survive and persist in the presence of a variety of pollutants such as heavy metals and organic chemicals. The genome of P. aeruginosa san ai is 6.98 Mbp long with a GC content of 66.08% and 6485 protein encoding genes. A large number of genes associated with proteins, responsible for microbial resistance to heavy metal ions andinvolved in catabolism of toxic aromatic organic compounds were identified. P.aeruginosa san ai is a highly cadmium-resistant strain. Proteome analysis of biomass after cadmium exposal confirmed a high tolerance to sublethal concentrations of cadmium (100mg/L), based on: extracellular biosorption, bioaccumulation, biofilm formation, controlled siderophore production and a pronounced metalloprotein synthesis. Proteins responsible for survival in osmostress conditions during exposure to elevated concentrations of cadmium (200mg/L) demonstrate a strong genetic potential of P. aeruginosa san ai for survival and adaptation. Sequencing of P. aeruginosa san ai genome provides valuable insights into the evolution and adaptation of this microbe to environmental extremes at the whole-genome level, as well as how to optimally use the strain in bioremediation of chemically polluted sites.

Cadmium specific proteomic responses of a highly resistant Pseudomonas aeruginosa san ai

AB  - Pseudomonas aeruginosa san ai is a promising candidate for bioremediation of cadmium pollution, as it resists a high concentration of up to 7.2 mM of cadmium. Leaving biomass of P. aeruginosa san ai exposed to cadmium has a large biosorption potential, implying its capacity to extract heavy metal from contaminated medium. In the present study, we investigated tolerance and accumulation of cadmium on protein level by shotgun proteomics approach based on liquid chromatography and tandem mass spectrometry coupled with bioinformatics to identify proteins. Size exclusion chromatography was used for protein prefractionation to preserve native forms of metalloproteins and protein complexes. Using this approach a total of 60 proteins were observed as up-regulated in cadmium-amended culture. Almost a third of the total numbers of up-regulated were metalloproteins. Particularly interesting are denitrification proteins which are over expressed but not active, suggesting their protective role in conditions of heavy metal exposure. P. aeruginosa san ai developed a complex mechanism to adapt to cadmium, based on: extracellular biosorption, bioaccumulation, the formation of biofilm, controlled siderophore production, enhanced respiration and modified protein profile. An increased abundance of proteins involved in: cell energy metabolism, including denitrification proteins; amino acid metabolism; cell motility and posttranslational modifications, primarily based on thiol-disulfide exchange, were observed. Enhanced oxygen consumption of biomass in cadmium-amended culture versus control was found. Our results signify that P. aeruginosa san ai is naturally well equipped to overcome and survive high doses of cadmium and, as such, has a great potential for application in bioremediation of cadmium polluted sites.

AB  - A comparative analysis of rhamnolipids from environmental isolates of Pseudomonas aeruginosa was undertaken to evaluate strain-specific rhamnolipid fingerprints obtained under different growth conditions. Environmental isolates of P. aeruginosa produced rhamnolipids on different types of substrates, including cheap and renewable sources like sunflower oil from deep fryers and sunflower oil mill effluent. Rhamnolipids were monitored by high-performance liquid chromatography-electrospray ionization interface mass spectrometry, which allowed fast and reliable identification and quantification of the congeners present. The highest concentration of total rhamnolipids of 3.33 g/l was obtained by the strain P. aeruginosa 67, recovered from petroleum contaminated soil, and strains D1 (1.73 g/l) and D2 (1.70 g/l), recovered from natural microbial consortia originated from mazut-contaminated soil, grown on sunflower oil as a carbon source. Di- to mono-rhamnolipids ratios were in the range of 0.90-5.39 for different media composition and from 1.12 to 4.17 for different producing strains. Rhamnolipid profiles of purified mixtures of all tested strains are similar with chain length from C8-C12, pronounced abundance of Rha-C10-C10 and Rha-Rha-C 10-C10 congeners, and a low content of 3-(3-hydroxyalkanoyloxy)-alkanoic acids. Concentrations of major congeners of RLs were found to slightly vary, depending on strain and growth conditions, while variations in minor congeners were more pronounced. Statistically significant increase of critical micelle concentration values was observed with lowering the ratio of total mono- to di-rhamnolipids ratio indicating that mono-rhamnolipids start to form micelles at lower concentration than di-rhamnolipids.

AB  - The production and characteristics of rhamnolipid biosurfactant obtained by the strain Pseudomonas aeruginosa san-ai were investigated. With regard to the carbon and nitrogen sources, several media were tested to enhance the production of rhamnolipids. Phosphate-limited proteose peptone-ammonium salt (PPAS) medium supplemented with sunflower oil as a source of carbon and mineral ammonium chloride and peptone as nitrogen sources greatly improved the production of rhamnolipid, from 0.15 on basic PPAS (C/N ratio 4.0) to 3 g L-1 on optimized PPAS medium (C/N ratio 7.7). Response surface methodology analysis was used for testing the effect of three factors, i.e., temperature, concentration of carbon and nitrogen source (mass %), in the optimized PPAS medium on the production of rhamnolipid. The isolated rhamnolipids were characterized by infrared (IR) spectroscopy and electrospray ionization mass spectrometry (ESI-MS). The IR spectra confirmed that the isolated compound corresponded to the rhamnolipid structure, whereas MS indicated that the isolated preparation was a mixture of mono-rhamno-mono-lipidic, mono-rhamno-di-lipidic and di-rhamno-di-lipidic congeners.
AB  - U ovom radu je opisana produkcija i karakterizacija ramnolipidnog biosurfaktanta soja Pseudomonas aeruginosa san-ai. Ispitan je uticaj različitih izvora ugljenika i azota na produkciju ramnolipida. Podloga proteoze pepton-amonijum soli sa ograničenom količinom fosfata (PPAS) sa suncokretovim uljem kao izvorom ugljenika i amonijum-hloridom i peptonom kao izvorom azota, znatno povećava produkciju ramnolipida, od 0,15, na PPAS sa standardnim sastavom (odnos C/N 4,0), do 3 g L-1 na optimizovanoj PPAS podlozi (odnos C/N 7,7). Metodom planiranog eksperimenta (response surface methodology) ispitan je efekat tri parametra: temperature, koncentracije izvora ugljenika i azota na optimizovanoj podlozi. Karakterizacija izolovanog preparata ramnolipida urađena je pomoću IR i ESI-MS analiza. IR analiza je potvrdila prisustvo strukturnih elemenata karakterističnih za ramnolipide. MS analiza pokazala je da reč o smeši u kojoj su prisutne mono-ramno-monolipidne, di-ramno-mono-lipidne i di-ramno-di-lipidne komponente.

abstract = "The production and characteristics of rhamnolipid biosurfactant obtained by the strain Pseudomonas aeruginosa san-ai were investigated. With regard to the carbon and nitrogen sources, several media were tested to enhance the production of rhamnolipids. Phosphate-limited proteose peptone-ammonium salt (PPAS) medium supplemented with sunflower oil as a source of carbon and mineral ammonium chloride and peptone as nitrogen sources greatly improved the production of rhamnolipid, from 0.15 on basic PPAS (C/N ratio 4.0) to 3 g L-1 on optimized PPAS medium (C/N ratio 7.7). Response surface methodology analysis was used for testing the effect of three factors, i.e., temperature, concentration of carbon and nitrogen source (mass %), in the optimized PPAS medium on the production of rhamnolipid. The isolated rhamnolipids were characterized by infrared (IR) spectroscopy and electrospray ionization mass spectrometry (ESI-MS). The IR spectra confirmed that the isolated compound corresponded to the rhamnolipid structure, whereas MS indicated that the isolated preparation was a mixture of mono-rhamno-mono-lipidic, mono-rhamno-di-lipidic and di-rhamno-di-lipidic congeners., U ovom radu je opisana produkcija i karakterizacija ramnolipidnog biosurfaktanta soja Pseudomonas aeruginosa san-ai. Ispitan je uticaj različitih izvora ugljenika i azota na produkciju ramnolipida. Podloga proteoze pepton-amonijum soli sa ograničenom količinom fosfata (PPAS) sa suncokretovim uljem kao izvorom ugljenika i amonijum-hloridom i peptonom kao izvorom azota, znatno povećava produkciju ramnolipida, od 0,15, na PPAS sa standardnim sastavom (odnos C/N 4,0), do 3 g L-1 na optimizovanoj PPAS podlozi (odnos C/N 7,7). Metodom planiranog eksperimenta (response surface methodology) ispitan je efekat tri parametra: temperature, koncentracije izvora ugljenika i azota na optimizovanoj podlozi. Karakterizacija izolovanog preparata ramnolipida urađena je pomoću IR i ESI-MS analiza. IR analiza je potvrdila prisustvo strukturnih elemenata karakterističnih za ramnolipide. MS analiza pokazala je da reč o smeši u kojoj su prisutne mono-ramno-monolipidne, di-ramno-mono-lipidne i di-ramno-di-lipidne komponente.",

AB  - The bioremediation of heavy metals using microorganisms has received a great deal of attention for its potential application in industry. Among the most common heavy metals used and the more widespread contaminants of the environment is mercury, cadmium, lead and copper. The mechanisms associated with metal removal by microorganisms are complex and occur as: biosorption of metal ions on the cell surface, intracellular uptake of metal ions or chemical transformation of metal ions by microorganisms. Biosorption is a passive uptake of metals based upon physicochemical interactions between metal and functional groups of the cell wall. Intracellular metal uptake, sometimes called active biosorption is slow and nutrient dependent process. This way of uptake is accumulation of metal cations by living cells, where they were modified and used for microbial needs or in form that is less/no toxic, and returned in environment. Various microbial species, mainly Pseudomonas, have been shown to be efficient in removal of the different heavy metals from polluted effluents, using combinations of mentioned mechanisms. This study represents trial to characterize and determine Cd2+ uptake by strain P. aeruginosa san ai. In order to evaluate tolerance of the strain, MIC for cadmium were determined, afterwards the metal uptake was monitored. Preliminary results showed that MIC for Cd was 2 mM. More than 80% of cadmium added to the culture, grown in submerged conditions, was found to be associated with biomass. Investigation of distribution or metal in cell fractions, as well in protein, lipid and polysaccharide fractions is underway.

Isolated rhamnolipid mixtures were analyzed by HPLC ESI-MS. Results showed that retention times depend on lipidic component of rhamnolipid, not only on molecular weight.

AB  - Zagadenje zivotne okoline teskim metalima je ozbiljan problem savremenog sveta. Među metalima koji su cesti zagadjivaci su: bakar, kadmijum i ziva. Cilj ovog rada je bilo ispitivanje osetljivosti soja Pseudomonas aeruginosa san ai na pomenute teske metale. Minimale inhibitome koncentracije kadmijuma i bakra od 2 i 4 mM, redom, pokazale su da je soj otporan na ova dva metala. MIK za zivu je bila najniza testirana koncentracija metala od 0,5 mM. Dobijene vrednosti MIK-aza kadmijum i bakar stavljaju soj P. aeruginosa san ai u grupu bakterija koje ispoljavaju visokutoleranciju prema ovim teskim metalima. Utvrdeno je daje ziva toksicna za soj. lspitivanja
mehanizama detoksikacije soja P. aeruginosa san ai u cilju potencijalne primene u procesima
bioremedijacije su u toku.
AB  - The pollution of the environment with toxic heavy metals is spreading throughout the world along with industrial progress. Among the common heavy metals used and the more widespread
contaminants of the environment are mercury, cadmium and copper. The present research was
performed in order to screen tolerance of strain Pseudomonas aeruginosa san ai to these heavy
metals Study showed that P. aeruginosa san ai is tolerant to cadmium and copper with MIC's of 2
and4 mM, respectively. MIC for mercury was the lowest tested concentration of 0.5 roM. This
suggests that P. aeruginosa san ai has a large potential for bioremediation of environments
contaminated with cadmium and copper and that mercury is toxic for the strain. In order to
improve possible application of strain, P. aeruginosa san ai in processes of bioremediation the
study of mechanisams of metal uptake are underway.
